摘 要:目的探讨生命早期Vit D缺乏及适量干预对大鼠肺发育的影响及可能机制。方法将18只成年雌性SD大鼠随机分为正常对照组(对照组)、Vit D缺乏组(缺乏组)、1,25-(OH)_2D_3干预组(干预组),每组6只。对照组及干预组予正常饲料、正常光照,缺乏组予不含Vit D饲料、避光,喂养2周后,与成年雄性SD大鼠交配,受孕第7日开始干预组予1,25-(OH)_2D_3(10μg·kg-1)隔天灌胃,对照组及缺乏组予1mL生理盐水代替,直到分娩后第21天,各组取新生第1、7、14、21天子鼠肺组织,光镜下观察肺形态结构,免疫组织化学方法检测肺SP-C蛋白水平表达。结果缺乏组子鼠较同日龄对照组肺组织肺泡数少,肺泡腔小,形态较不规则,肺泡间隔较宽;干预组子鼠较同日龄对照组肺泡数稍增加,肺泡腔大小均匀,肺泡间隔较薄;3组子鼠随着日龄增加,SP-C的表达量逐渐增多,缺乏组子鼠SP-C表达量均低于同日龄对照组及干预组,差异有统计学意义(P<0.05);干预组子鼠SP-C表达量均高于对照组,其中第14、21天比较差异有统计学意义(P<0.05)。结论维生素D在生命早期可影响大鼠肺发育,适量补充1,25-(OH)_2D_3有助于大鼠肺发育,其机制可能与影响肺SP-C的表达有关。Objective To explore the effects of vitamin(vit)D deficiency and appropriate intervention in early life on lung development in rats and its possible mechanisms of action.MethodsEighteen adult female Sprague-Dawley rats were randomly divided into three groups,with 6rats in each group.The normal control group(control group)and 1,25-(OH)2D3intervention group(intervention group)were given normal diet and light.The Vit D deficiency group(deficiency group)was fed Vit D-free diet and kept away from light.After treatment for 2weeks,female rats copulated with adult male rats.From the 7th day of pregnancy to the 21 st day after delivery,the intervention group was intragastrically administered 1,25(OH)2D3(10μg·kg^-1 every other day.The control group and deficiency group were given normal saline(1ml)by gavage.Lung tissue of newborns was obtained on postnatal days 1,7,14 and 21.Lung morphology was observed under light microscope,and SP-C protein expression was detected by immunohistochemical staining.Results Compared with control group,lung tissue of rats in deficiency group had decreased alveolar number and reduced alveolar cavity with irregular morphology and thickened alveolar septum,but lung tissue of rats in intervention group had increased alveolar number with uniform alveolar size and thin alveolar septum.The SP-C expression in newborn rat lung tissue increased with age in all the three groups.However,SP-C expression in deficiency group was significantly lower than that in control group and intervention group(P〈0.05).Moreover,SP-C expression in intervention group was higher than that in control group,and the difference was significant on the14 th and 21 st days(P〈0.05).Conclusion Vit D can affect lung development in early life in rats.Proper supplement of 1,25-(OH)2D3 is helpful for the development of rat lung,and the mechanism may be related to the expression of SP-C.
