摘 要:为研究不同剂量LiCl对绒山羊毛囊生长和β-连环蛋白(β-catenin)m RNA表达的影响,将分离的绒山羊初级毛囊随机分为5组,每组24个重复,对照组为William'E基础培养液。试验组在基础培养液中依次添加2,5,10和20mmol·L^(-1)的LiCl,培养期为7d,每日测量毛囊生长长度。运用荧光定量PCR技术测定各组β-cateninm RNA表达水平。结果表明:2~10 mmol·L^(-1)LiCl组的β-catenin m RNA表达水平和生长长度极显著高于对照组(p<0.01),毛囊形态维持良好;LiCl浓度为20mmol·L^(-1)组的β-catenin m RNA表达量及生长长度极显著低于对照组(p<0.01),体外培养毛囊生长受到抑制。添加2~10mmol·L^(-1)的LiCl可促进绒山羊毛囊生长,促使β-catenin m RNA表达水平提高,其中5mmol·L^(-1)为较优浓度。通过本试验研究发现,适量的LiCl在一定程度上有利于初级毛囊的生长及形态的维持,促进了β-catenin m RNA表达。This experiment was to study in the hair follicle beta catenin (β- catenin) mRNA expression and the growth of hair follicle under the effect of different doses of LiCl. The cashmere goat primary follicles which have been separated were randomly divided into 5 groups. Each group has 24 replicates in this study. The control group of this study was William E basal medium. In the experimental group, LiCl was added in the order of 2, 5, 10 and 20 mmol·L-1, based on the basal medium. This training lasted for 7 days. The hair follicles' growth length was measured every day. Real-time PCR technology was used to determine the level of β-catenin mRNA expression in each group of hair follicle. The results showed that the expression level of β-catenin mRNA and the hair follicle's growth length were significantly higher (p 〈0.01) than the control group in the 2 to 10 mmol·L-1 LiCl groups. The shape of hair follicles maintained good in this group. The expression level of β-caten in mRNA and the hair follicle's growth length were significantly lower (p〈0.01) than the control group in the 20 mmol·L-1 LiCl group. The hair follicle's growth length were inhibited in vitro. In conclusion, the adjunction of 2 to10 mmol·L-1 LiCl could promote hair follicle growth and the level of β-catenin mRNAs' expression, in which 5 mmol·L-1 has the best effect. Through this experimental study: a bit of LiCl could promote the growth of the primary follicles and maintain primary follicles of the form promoted the expression of β-catenin mRNA.
