三叶青黄酮对结肠癌SW620细胞增殖凋亡的影响

摘　　要：[目的]观察三叶青黄酮对结肠癌SW620细胞增殖凋亡的作用并探讨可能的机制。[方法]采用体外细胞培养技术,MTT法观察三叶青黄酮对SW620细胞增殖的影响,流式细胞术分析细胞凋亡率,Western Blotting检测Cle-caspase3、Cle-caspase9、Bax、Bcl-2蛋白表达改变。[结果]三叶青黄酮能显著地抑制SW620细胞增殖,并且具有剂量和时间依赖性。三叶青黄酮能显著地促进SW620细胞凋亡,并且呈剂量依赖性上调Cle-caspase3、Clecaspase9、Bax表达,下调Bcl-2表达。[结论]三叶青黄酮对人结肠癌SW620细胞具有抑制增殖和促进凋亡的作用。[Objective]To observe the effect and possible mechanism of Radix Tetrastigma Hemsleyani Flavone（RTHF）on the proliferation and apoptosis of human colon cancer SW620 cells.[Methods]Human colon cancer cell line SW620 was cultured in vitro and treated with different concentrations of RTHF.MTT method was used to observe the effect of RTHF on the proliferation of SW620 cells.The apoptosis rate was analyzed by flow cytometry.The expression changes of Cle-caspase3,Cle-caspase9,Bax and Bcl-2were detected by Western Blotting.[Results]RTHF could significantly inhibit the proliferation of SW620 cells with dose and time dependent manner.RTHF could significantly promote the apoptosis of SW620 cells.RTHF could increase the expression of Cle-caspase3,Cle-caspase9 and Bax,and down-regulate the expression of Bcl-2in a dose dependent manner.[Conclusion]RTHF can inhibit the proliferation of human colon cancer SW620 cells,and promote apoptosis of SW620 cells.
