摘　　要：为了保证胚胎移植的效果,达到良种繁育的目的,对胚胎移植的受体牛进行了牛病毒性腹泻病毒(BVDV)-持续感染(PI)牛的筛查。首先采取受体牛血清样本,然后提取样本RNA,反转录为c DNA,进行RT-巢式PCR,根据琼脂糖凝胶电泳结果,确定受体牛是否是BVDV-PI牛。初次检测结果为阴性的牛,可作为胚胎移植的备选牛。初检阳性的牛,进行隔离,3周后进行复检,连续进行两次复检,如果3次检测结果都为阳性,认定该牛为BVDV-PI牛,予以淘汰。如果复检为阴性,则判为BVDV急性感染牛,可继续使用。结果 108头受体牛中,确认3头受体牛为BVDV-PI牛,予以淘汰,BVDV-PI牛阳性率2.8%。剩下的105头受体牛,通过直肠检查可用于胚胎移植的81头。45天后孕检,怀孕61头,受胎率75.3%。In order to ensure the effect of embryo transfer and improve breeding,recipient cows were screened to determine whether they are BVDV-PI cattle.First,recipient cows serum samples were collected,and then RNA was extracted,and used for BVDV analysis by RT-nested PCR.If the first test result of the recipient cows is negative,they can be used as an alternative cow for embryo transfer.Early detection of positive cows were isolated and rechecked after three weeks,and were then rechecked twice in a row.If the three test results were positive,the cow was identified as BVDV-PI cattle,and was eliminated.If the recheck was negative,the cow could be judged as an acute BVDV infection,and could be used.Three cows from 108 recipient cows were identified as BVDV-PI cattle,and were eliminated.BVDV-PI cattle positive rate of 2.8 %.The remaining 105 recipient cows could be used for embryo transfer.And then,81 cows were confirmed to be used for embyo transfer by rectal examination.After 45 days,pregnancy 61 were examined to be pregnant,and then pregnancy rate was 75.3 %.
