摘  要:目的:探讨云南汉族人群低相对分子量蛋白酶体(LMP)2基因单核苷酸多态性(SNP)与非小细胞肺癌的相关性。方法:选择云南汉族非小细胞肺癌患者467人(病例组)和健康正常对照490人(对照组),对病例组患者进行病例分型和临床分期,采用TaqMan探针基因分型法对所有对象进行LMP2基因的3个SNPs(rs1351383、rs17587和rs2127675)位点基因分型,用卡方检验比较病例和对照组间3个SNP等位基因和基因型的分布差异,用Haploview软件构件这3个SNP的个体单倍型,并分析两组间及不同病理类型肺癌患者与对照间的单倍型分布差异。结果:3个SNP位点的等位基因及基因型在病例组和对照组中的分布差异均无统计学意义(P>0.05),但rs1351383位点的C等位基因在鳞癌组中的频率显著高于对照组,差异具有统计学意义(P=0.049,OR=0.769,95%CI:0.591~0.999);单倍型rs1351383-rs17587-rs2127675的G-C-G在病例组中(OR=1.461,95%CI:1.075~1.984)以及肺腺癌患者组(OR=1.608,95%CI:1.142~2.265)中的分布显著高于对照组,单倍型rs1351383-rs17587-rs2127675的G-C-A在总病例组中(OR=0.593,95%CI:0.422~0.834)以及肺腺癌患者组(OR=0.541,95%CI:0.365~20.823)中的分布显著低于对照组。结论:LMP2基因的3个SNPs(rs1351383、rs17587和rs2127675)单个位点与云南汉族人群肺癌无相关性,但3个SNPs构建的单倍型可能是云南汉族人群肺癌患病的风险因子。Objective: To investigate the correlation of single nucleotide polymorphisms (SNP) of low molecular weight proteasome (LMP) 2 gene with non-small cell lung cancer in Han nationality of Yunnan province. Methods: TaqMan assay was used to genotype 3 SNPs (rs1351383, rs17587, and rs2127675) in LMP2 gene in 467 non-small cell lung cancer patients (case group) and 490 healthy people (control group) of Han nationality in Yunnan province. The distribution difference of allele and genotypes of the 3 SNPs between the 2 groups was analyzed with chi-square test, and individual haplotypes of the 3 SNPs were constructed with Haploview software. The haplotype distribution differences between the 2 groups, and between lung cancer patients of different pathological types in control group were analyzed. Results: The allelic and genotype distributions of the 3 SNPs of the 2 groups were not significantly different(P 〉0.05). While the rs1351383-C mutant was significantly high in squamous cell carcinoma subgroup (P =0. 049, OR =0. 769, 95% CI: 0. 591 - 0.999) than in control group. And there were notable differences of genotype distribution in some haplotypes. The distributions of haplotype rs1351383-rs17587-rs2127675 : G-C-G in total case group ( OR = 1. 461,95% CI: 1. 075 1. 984 ) and in lung adenocarcinoma group ( OR = 1. 608, 95% CI: 1. 142 - 2. 265 ) were significantly higher than in control group; oppositely, the distribution of haplotype rs1351383-rs17587-rs2127675 : G-C-A in total case group ( OR = 0. 593, 95 % CI: 0.422 - 0. 834) and in lung adenocarcinoma group ( OR = 0. 541, 95% CI: 0. 365 - 20. 823 ) were significantly lower than in control group. Conclnsions: Individual of the 3 SNPs in LMP2 gene has not correlation with the incidence of lung cancer sig- nificantly in Han nationality of Yunnan province. But the haplotypes constructed by the 3 SNPs might be a risk factor of lung cancer in the population.
