摘 要:目的观察熄风静宁颗粒对Tourette综合征(TS)模型大鼠脑纹状体多巴胺D2受体(DRD2)和多巴胺转运蛋白(DAT)mRNA表达的影响。方法 Wistar雄性大鼠36只随机分为空白对照组、模型组、氟哌啶醇组、熄风静宁颗粒低剂量组、中剂量组、高剂量组,采用亚氨基二丙腈(IDPN)腹腔注射建立TS大鼠模型,造模后连续给药6周,采用RT–PCR法检测大鼠脑纹状体中DRD2和DAT mRNA表达的情况。结果 IDPN诱导的模型大鼠的体质量显著低于正常大鼠(P<0.01);给药前后熄风静宁颗粒中剂量组TS模型大鼠体质量增长与正常大鼠体质量的增长无差异;TS模型大鼠脑纹状体DRD2 mRNA与正常组比较呈现高表达(P<0.01);熄风静宁颗粒中、高剂量组和氟哌啶醇可降低TS模型大鼠脑纹状体DRD2 mRNA的表达(P<0.05);TS模型大鼠脑纹状体DAT mRNA表达与正常组比较显著增高(P<0.01);中药颗粒高剂量组能够降低TS模型大鼠脑纹状体DAT mRNA的表达,但氟哌啶醇作用更明显(P<0.05)。结论熄风静宁颗粒尚具有部分调节机体状态的作用;Tourette综合征存在多巴胺功能的亢进,熄风静宁颗粒能够通过降DRD2受体的敏感性,抑制多巴系统系统功能的亢进而发挥药物治疗的作用。Objective: To investigate the mRNA expression of dopamine receptor D2 (DRD2) and dopamine transporter(DAT) in rat brain striatum of tourette syndrome model. Methods: Male Wistar rats (n = 36) were randomly divided into 6 groups: control group,model group,haloperidol group,low-dose of Xifeng Jingning granule group, middle-dose of Xifeng Jingning granule group and high-dose of Xifeng Jingning granule group. The tourette syndrome model was established in rats by intraperitoneal injection of iminodipropionitrile (IDPN). After modeling the rats were given medicines for 6 weeks. The mRNA expression of dopamine receptor D2(DRD2) and dopamine transporter(DAT) in rat brain striatum of Tourette Syndrome model were investigated by using real-time quantitative polymerase chain reaction (RT-PCR). Results: The weight of tourette syndrome rats by intraperitoneal injec- tion of iminodipropionitrile was significantly lower than normal rats (P 〈 0.01). There was no difference on rat weight gain between middle-dose of Xifeng Jingning granule group and control group (P〉 0.05). Compared with control group,dopamine receptor D2 mRNA in striatum of tourette syndrome rats showed higher expression (P 〈 0.01 ). Middle-dose,high-dose of Xifeng Jingning granule group and haloperidol group could reduce the expression of dopamine receptor D2 mRNA in tourette syndrome rat striatum (P〈 0.05). Compared with control group, dopamine transporter mRNA in tourette syndrome rat striatum showed higher expression (P 〈 0.01). Dopamine transporter mRNA expression in tourette syndrome rat striatum in high-dose of Xifeng Jingning granule group and haloperidol group could get reduced,but more obviously in haloperidol group (P〈 0.01 ). Conclusion: Xifeng Jingning granule has a partial role in regulating body state;Tourette syndrome may induce overactivity of dopamine system.
