风湿痹痛方治疗大鼠胶原性关节炎的实验研究

摘　　要：目的探讨风湿痹痛方对类风湿关节炎的治疗作用及其初步机制。方法采用大鼠足部皮内注射牛Ⅱ型胶原乳化剂,建立胶原诱导的关节炎模型(CIA),造模成功后设空白对照组、模型对照组、雷公藤多苷组(7.5 mg/kg)及风湿痹痛方(9.5,19,38 g/kg)3个剂量组,连续给药14 d。观察风湿痹痛方对CIA大鼠足肿胀的治疗作用。ELISA法检测血清肿瘤坏死因子-α(TNF-α)、sFas、sFasL含量的变化;Western Blot法检测CIA大鼠滑膜组织TNF-α、IL-1β、半胱氨酸蛋白酶-8(Caspase-8)和FLIP的表达。结果造模14 d后,模型大鼠与正常对照组相比足肿胀明显,风湿痹痛方可明显减轻CIA大鼠的足肿胀度。与模型组相比,风湿痹痛方高剂量组大鼠血清中sFas、sFasL含量明显降低(P<0.05);大鼠滑膜组织Caspase-8表达含量显著升高(P<0.05)。结论风湿痹痛方对类风湿关节炎具有免疫调节作用,其机制可能与提高Caspase-8的表达同时影响sFas/sFasL介导的滑膜细胞凋亡,促进细胞凋亡,减少关节损伤有关。Objective： To discuss the therapeutic effects and preliminary mechanism of Fengshi Bitong（FSBT） prescription on treatment of rheumatoid arthritis （RA）. Methods： Rats were given foot intradermal injection of bovine type collagen emulsion to establish collagen-induced arthritis （CIA） model. After successful modelling, they were randomly divided into 6 groups： normal control group;CIA model group ;tripterygium glycosides group （7.5 mg/kg）;FSBT groups（9.5,19,38 g/kg）,and were continuous administered for 14 days. The effect of FSBT on paw edema was observed. Serum levels of TNF-α, sFas, sFasL were detected by ELISA. Expression of TNF-α, IL- 1β, caspase-8 and FLIP of CIA rats＇ synovial tissue were detected by Western Blot. Results： In model group,the paw edema was significant compared with normal control group, and FSBT could significantly reduce the paw edema. The serum levels of sFas,sFasL in FSBT high dose group （38 g/kg） were significantly decreased （P〈 0.05） compared with CIA model group. Expression of caspase-8 was significantly increased （P 〈 0.05）. Conclusion： FSBT may have immune regulation effects on rheumatoid arthritis. Its mechanism may be related to increasing the expression of caspase-8 and affecting apoptosis of synovial cell,which is mediated by sFas/sFasL.
