摘 要:目的探讨白芍总甙在脓毒症模型中的抗炎效果及作用途径。方法60只SD大鼠以盲肠结扎手术制作脓毒血症模型,均分为假手术组、模型组及白芍总甙组。分别检测各临床数值(WBC、Pa02、AST、ALT及Scr),采用ELISA法检测各组血清IL-1、IL-6及TNF-α水平,RT—PCR定量检测各组肺组织中TLR-4mRNA及NF-kBp65mRNA的表达,同时比较各组湿/干肺比值。结果与模型组相比较,假手术组所测各临床数值,血清IL-1、IL-6及TNF-α水平明显降低,肺组织中TLR-4mRNA及NF-kBp65mRNA的表达均明显降低,各组差异有显著性(P〈0.05)。结论TLR—s/NF—kB信号途径参与了脓毒血症模型的致病过程,白芍总甙能通过调节该信号通路发挥下调IL-1、IL-6及TNF-α水平,减轻脓毒症的炎症反应及肺组织的损伤。Objective To investigate the effects of total glucosides of paeomy(TGP)on sepsis by CLP in SD rats and explore pathway involved. Methods Sepsis was induced in 60 rats by CLP. The model rats were divided into three groups(n=20) :No CLP group, model control group(receiving 0.9 % NaCI solution), TGP treatment group. The basic clinical data value were examined, the serum levels of interleukin-1 (IL- 1), IL-6 and TNF-a were determined by enzyme-linked immunosorbant assay(ELISA), and the lung expression of Toll like receptor-4 and Neural transfer factor Bp65 were determined by RT-PCR. Lastly, the ratio of wet to dry of lung was recorded. Results Compared with the model control group, each of clinical index(WBC, PaO2, AST, ALT and Scr) was tested decreased significantly in TGP group. There is difference among them(P〈0.05) ; the serum levels of IL-1, IL-6 and TNF-a also declined significantly, the expression of TLR-4mRNA and NF-kBp65mRNA in lung tissue decreased obviously, Significant differences were found in the above parameters between TGP treatment group and the model group(P 〈0.05). It is significant difference for ratio of wet lung to dry lung between TGP and model group(P〈0. 05). Conclusion The pathway of TLRs/NF-kB has been proved to take part in the course of sepsis model, TGP can alleviate inflammatory reaction and organs damage in sepsis by modeling the pathway of TLRs/NF-kB.
