摘 要:目的建立一种测定血清中1,5-脱水葡萄糖醇(1,5-AG)的方法,辅助诊断糖尿病。方法糖尿病患者(糖尿病组)与健康体检人员(健康对照组)各100例,采用葡萄糖激酶去除血清样品内源葡萄糖干扰,过氧化氢(H_2O_2)在4-氨基替比林(4-AAP)、过氧化物酶(POD)的存在下,可生成红色醌亚胺化合物,发生显色反应进行比色测定。结果该方法最低检测限为1.98μmol/L,1,5-脱水葡萄糖醇在663μmol/L以内线性良好。测定100例糖尿病组患者血清1,5-脱水葡萄糖醇含量为(30.7±16.4)μmol/L,显著低于健康对照组的(170.6±48.2)μmol/L,差异有统计学意义(P<0.05)。结论该法简便快速、灵敏、准确和稳定、特异,可用于糖尿病的及时诊断与监控,适合于各类实验室常规应用。Objective To establish determination method for serum 1,5- anhydroglucitol holoenzyme(1,5-AG) to assist diagnosis of diabetes mellitus.Methods There were diabetes mellitus patients(diabetes mellitus group) and healthy people(healthy control group),with 100 cases in each group.Glucokinase was applied to eliminate interference by glucose in serum samples.Chromogenic reaction of by hydrogen peroxide(H_2O_2) generating red quinone- imine compound in 4- amino- antipyrine(4-AAP) and peroxidase(POD) were used in colorimetric determination.Results The lowest detectable limit of this method was 1.98 μmol/L,and1,5- anhydroglucitol holoenzyme showed good linearity within 663 μmol/L Serum 1,5- anhydroglucitol holoenzyme content was(30.7± 16.4) μmol/L in 100 diabetes mellitus patients,which were obviously lower than(170.6± 48.2) μmol/L in the control group,and their difference had statistical significance(P〈0.05).Conclusion As an easy,convenient,sensible,accurate,stable and specific method,it can be applied in timely diagnosis and monitoring of diabetes mellitus.This method is suitable for laboratory conventional application.
