摘 要:目的测定甘草次酸改性前后的表观油水分配系数,考察其水溶性改善情况。方法建立紫外分光光度法,采用摇瓶法(正辛醇-水体系)测定甘草次酸及聚乙二醇单甲醚改性甘草次酸在不同油水比例以及不同p H值下的表观油水分配系数(lg P)。结果甘草次酸在无水乙醇中的最大紫外吸收波长为248nm,线性回归方程为A=0.0233C-0.0008(n=6,r=0.9999),在6.0~36mg/L范围内,线性关系良好。改性后甘草次酸的表观油/水分配系数下降到-0.538^-0.767。结论难溶性中药有效成分甘草次酸经聚乙二醇单甲醚改性后在水中的溶解性显著提高。Objective To determine the apparent oil/water partition eoefficient of insoluble glycyrrhetinic acid(GA) before and after the modification and observe itswater solubility. Methods UV spectrophotometry was estab- lished to determine the content of GA, and the apparent oil/water partition coefficient(lgP) was determined by shaking flask method in different pH solutions and different proportions of oil and water. Results The linear regression equation in ethanol was A = 0. 0233C - 0. 0008 ( n = 6, r = 0. 9999 ) at 248nm, and linear relationship was excellent in the range of 6 - 36mg/L. The lgP of modified GA, which was methoxypolyethylene glycols-glycyrrhetinie acid (mPEG-GA) was al- readydecreased to -0. 538 - -0. 767. Conclusion The solubility of the effective components of Chinese herbal medi- cine in the water could be significantly increased.
