摘  要:目的探究CD26表面分子在正常皮肤组织和瘢痕疙瘩成纤维细胞中表达的差异。方法正常皮肤成纤维细胞(NFs)组的正常皮肤组织取自接受腹部整形术的10例患者;瘢痕疙瘩成纤维细胞(KFs)组瘢痕疙瘩组织取自此前未经过任何治疗的10例瘢痕疙瘩患者的胸部和腹部。利用酶消化法获得原代KFs和NFs,应用流式分析技术检测CD26表面分子在2组细胞中的表达差异。用流式分选技术对NFs和KFs组中CD26表达阳性(CD26+)与表达阴性(CD26-)细胞进行分选,体外常规培养4 d后对细胞形态进行初步分析。结果 CD26表面分子在KFs和NFs中均可表达,但KFs组表达率(46.65±13.97)%明显高于NFs组(24.75±16.85)%,差异具有统计学意义(P<0.01)。经流式分选后,CD26+与CD26-细胞群在NFs与KFs组中形态均无明显差异。结论 CD26分子在瘢痕疙瘩组织成纤维细胞中的表达率明显高于正常皮肤组织成纤维细胞中的表达率,CD26-和CK26+细胞群在形态上无明显差异。Objective To analyze the expression of the CD26 marker on normal skin fibroblasts (NFs) and keloid fibroblasts (KFs). Methods Normal skin tissues in the NFs group were obtained from individuals who underwent abdominal plastic resection. Keloid tissues in the breast and abdomen were harvested in surgery from patients who did not receive any treatment for their keloids. Then NFs and KFs were obtained through the enzyme digestion method. Flow eytometry technology was used to analyze the expression of the CD26 marker on NFs and KFs, and isolate the CD26 positive expressed (CD26+) and CD26 negative expressed (CD26-) cell population of NFs and KFs. Then the cell morphology of CD26-/+ fibroblasts were observed after 4 days in vitro culture by inveried microscope. Results The CD26 marker was expressed in both KFs and NFs. KFs exhibited a higher expression rate of CD26 (46.65 ±13.97)% than NFs with that of (24.75 ±16.85)% (P〈0.01). Observation of cell morphology after 4 days in vitro culture exhibited similar shape and cell density between CD26- cell population and CD26+ cell population in both the NFs and KFs groups. Conclusion KFs exhibited a higher expression rate of CD26 than NFs, while the CD26- and CD26+ cell population in both the NFs and KFs groups showed similar morphological characteristics.
