摘  要:以野扁桃为实验材料,提取叶片的RNA,设计基因特异性引物,利用RT-PCR克隆转录因子CBF的c DNA全长,命名为Als CBF1-A(登录号KU975456);并对其基因和蛋白进行了生物信息学分析。结果表明,该转录因子基因的CDS区长699 bp,编码232个氨基酸,编码蛋白相对分子质量约为26.027 2 k D,p I 4.84;由无规则卷曲(C),α-螺旋(H),β-折叠(E)组成,分别占52.17%,21.74%和26.09%;有较强的亲水性,无跨膜区和信号肽存在,有8处丝氨酸磷酸化位点、2处苏氨酸磷酸化位点、2处酪氨酸磷酸化位点;同源性分析表明Als CBF1-A基因在高等植物中保守性较高。本研究显示CBF转录因子可能与野扁桃的抗寒性相关,此研究为野扁桃抗寒分子机理的进一步研究提供了帮助。Amygdalus ledebouriana Schleche were used as the experiment materials in this research, RNA was extracted, the full-length’s c DNA of plant transcription factor CBF named Als CBF1-A(Accession number: KU-975456) was cloned by RT-PCR from Amygdalus ledebouriana leaves. The bioinformatics analysis of gene and protein was carried out. The results showed that the region of the transcription factor gene CDS was 699 bp, and it encoded 232 amino acids. The relative molecular weight of the encoded protein was about 26.027 2 k D, p I 4.84. It was consist of non-regular curls(C), alpha helices(H) and beta-fold(E), which were respectively accounted for52.17%, 21.74%, and 26.09%. It had strong hydrophilic property, but no transmembrane domain and signal peptide, also there were 8 serine phosphorylation sites, 2 threonine phosphorylation sites, 2 tyrosine phosphorylation sites. Homology analysis showed that the Als CBF1-A gene was highly conserved in higher plants. The research showed that Als CBF1-A transcription factor may be related to the cold resistance of wild almond. This would be useful for the further study on the molecular mechanism of cold resistance of wild almond.
