摘 要:在前期巴西橡胶树胶乳转录组测序的基础上,通过PCR克隆了1个编码巴西橡胶树MADS-box转录因子的基因,命名为Hb MADS5。Hb MADS5全长1 077 bp,编码358个氨基酸,推测Hb MADS5蛋白分子量为40.75 k D,等电点为6.38。氨基酸序列比对分析表明,Hb MADS5具有MADS-box家族典型的MADS-box保守结构域,与蓖麻、葡萄、麻风树、甜橙、木本棉、蒺藜状苜蓿等物种的MADS-box家族成员具有较高的同源性,分别为79%、74%、72%、70%、65%和59%。Hb MADS5与麻风树中的1个MADS-box成员亲缘关系最近。Hb MADS5在巴西橡胶树根、茎、叶、花和胶乳等中都有表达,其中在胶乳中表达量最高,在根中表达量最低;茉莉酸甲酯和乙烯能诱导Hb MADS5的表达,茉莉酸甲酯处理12 h后Hb MADS5表达量提高了10倍,乙烯处理48 h后Hb MADS5表达量提高了2倍。本研究将为进一步研究Hb MADS5在巴西橡胶树中的功能提供了帮助。Based on the transcriptome sequencing of Hevea brasiliensis, a new gene coding for MADS-box transcription factor, designated as Hb MADS5, was isolated from Hevea brasiliensis by PCR. Hb MADS5 consisted of a 1 077 bp open reading frame encoding 358 amino acids with molecular mass of 40.75 k D and p I 6.38. Hb MADS5 contained a typical conserved MADS-box motif at the N terminus. The deduced amino acid sequence of Hb MADS5 showed high identity of 79%, 74%, 72%, 65% and 59% to those of the MADS-box proteins from Ricinus communis, Vitis vinifera, Jatropha curcas, Citrus sinensis, Gossypium arboreum and Medicago truncatula.The phylogenetic analyses showed that Hb MADS5 is closest to a member of the MADS-box that from Jatropha curcas. Hb MADS5 was expressed at different levels, with the highest transcription in the latex and lowest in the roots. The expression of Hb MADS5 was induced by jasmonic acid and ethylene, increased 10-fold after 12 h treated by jasmonic acid and increased 2-fold after 48 h treated by ethylene respectively. Characterization of Hb MADS5 will attribute to understand their possible function in rubber tree.
