摘  要:建立杂交兰无菌播种和再生体系,获得一定数量的杂交后代群体,为新品种的选育提供基础材料。以大花蕙兰‘肯尼’为母本和墨兰‘太平洋’进行种间远缘杂交,取其成熟种子进行无菌播种,探讨不同基本培养基、植物生长调节剂和添加物对其种子萌发、根状茎增殖与分化及壮苗生根等的影响。结果表明:种子萌发最适配方为:1/2MS+6-BA 1.5 mg/L+NAA 0.5 mg/L+活性炭1.0 g/L;根状茎增殖与分化培养基最适配方为1/2MS+6-BA 2.0 mg/L+NAA 0.5 mg/L,培养70天分化成苗率达88.6%。适宜的壮苗生根培养基为MS+6-BA 1.0 mg/L+NAA 2.0 mg/L+香蕉泥90 g/L+活性炭2.0 g/L。室外闭瓶炼苗30~40天,再开盖炼苗3~5天后,以发酵的树皮加碎石子为基质,60天左右种苗成活率达98.62%。Aseptic seeding and the establishment of regeneration system play an important role in cross-breeding of orchid, obtaining a certain number of hybrid progeny population, and providing basic materials for the breeding of new varieties. An aseptic seeding propagation for interspecific hybrid between Cymbidium hybridum‘Ken Ni'and C. sinense‘Pacific Ocean'was established. Different minimal medium, hormone andadditives were designed to study the effects on the germination, rhizome proliferation and differentiation andseedling root. The results showed that 1/2MS+ 6-BA 1.5 mg/L+ NAA 0.5 mg/L+ AC 1.0 g/L was the suitablemedium for the hybrid seed germination, 1/2MS+ 6-BA 2.0 mg/L+ NAA 0.5 mg/L was the ideal medium for therhizome proliferation and differentiation, the differentiation rate was 88.6% after culturing for 70 days. MS+ 6-BA 1.0 mg/L+ NAA 2.0 mg/L+ banana 90 g/L+ AC 2.0 g/L was the right medium for inducing the root growth.Test- tube train seedling 30- 40 days in closed bottle and opening lid after 3- 5 days in greenhouse,fermentation bark and blinding were best media for temporary planting of tissue culture seedling. About 60 days, the seedling survival rate was 98.62%.
