湿热清对寻常型银屑病外周血单个核细胞Ets-1 mRNA水平的影响

摘　　要：目的 :观察湿热清对寻常型银屑病血热证患者外周血单个核细胞Ets-1 mRNA水平的影响,探讨转录因子Ets-1与寻常型银屑病血热证的相关性,揭示湿热清加味治疗的微观作用机制。方法:采用实时定量PCR法检测28例寻常型银屑病血热证患者与20例健康者外周血单个核细胞中Ets-1 mRNA水平,观察患者经湿热清加味治疗后Ets-1 mRNA水平的变化。结果:寻常型银屑病血热证患者外周血中Ets-1 mRNA水平显著高于健康对照组(P<0.01),给予湿热清加味治疗后Ets-1mRNA表达量明显降低(P<0.05)。结论:寻常型银屑病血热证患者外周血中Ets-1 mRNA水平明显高于正常人,湿热清加味治疗后明显降低,提示湿热清加味可能通过调节转录因子Ets-1表达量间接影响T淋巴细胞分化,从而对寻常型银屑病血热证患者发挥治疗作用。Objective：To observe the effect of Shireqing on the mRNA level of peripheral blood mononuclear cell（PBMC） in the patients with psoriasis vulgaris,to study the correlation between transcription factor Ets-1and psoriasis vulgaris of blood heat type,and to reveal the microscopic mechanismof modified Shireqing.Method：Levels of PBMC ETs-1 mRNA of 28 patients with psoriasis vulgaris of blood heat type and 20 healthy people were detected by real-time quantitative PCR,and the changes on levels of Ets-1 mRNA before and after treatment with modified Shireqing. Result：The level of PBMC Ets-1 mRNA in 28 patients with psoriasis of blood heat type were significantly higher than those in the healthy people（P〈0.01）. The expression level of Ets-1 mRNA was significantly reduced after the treatment with modified Shireqing（P〈0.05）. Conclusion：The PBMC Ets-1 mRNA level in patients with blood heat syndrome of psoriasis vulgaris was significantly higher than that of normal people,which could be significantly reduced after treated with modified Shireqing,suggesting that modified Shireqing may regulate the expression level of transcription factor Ets-1 to indirectly affect T lymphocyte differentiation. Through this mechanism,Shireqing plays a role in the treatment of patients with psoriasis vulgaris of blood heat type.
