摘  要:目的:研究蓬子菜中总黄酮和木犀草苷对脂多糖(LPS)诱导的脐静脉内皮细胞(HUVEC)炎症损伤的保护作用。方法:以HUVEC为研究对象,用CCK-8检测总黄酮及木犀草苷对HUVEC增殖的影响;采用流式细胞仪检测不同浓度LPS对HUVEC细胞周期的影响;酶联免疫分析法检测总黄酮及木犀草苷对LPS诱导的HUVEC炎症因子生成的影响;实时定量PCR法检测HUVEC中P-选择素E-选择素mRNA水平。Western blot法测NF-κB,P-IκB蛋白表达。结果:药物浓度在6.25、12.5、25μg/m L时细胞增殖率最大;总黄酮及木犀草苷能明显下调LPS诱导的炎症因子和黏附分子的分泌,同时也降低了P-选择素E-选择素的表达,NF-κB,P-IκB蛋白表达量。结论:LPS对HUVEC有抑制作用呈剂量、时间依赖性,给予总黄酮和木犀草苷后,损伤有明显改善。Objective: To observe the protective effect of galuteotin and flavonoids from Galium verum L. on HUVEC inflammatory injury induced by LPS. Methods: The method called CCK - 8 was used to detect the influence of drugs on the proliferation of HUVEC. The influences of different LPS concentrations on the cell cycle were compared by FCM (flow eytometry). Use ELISA to detect the influence of LPS on the generation of inflammation factor after giving drugs. The expression levels of E - selection and P - selection mRNA were measured by RT - PCR. Western blot was used to detect the protein expression of NF - κB, p - Iκ - B. Results : When the concentrations of drugs were 6.25, 12.5, 25 μg/mL respectively, the proliferation rate was the best. The drugs had an obvious effect on decreasing the secretion of the inflammation factor in the HUVEC induced by LPS. Meanwhile, they could decrease the relative expression of E - selection and P - selection and the protein expression of NF - κB, P - IκB. Conclusion : The inhibitive effect of LPS presents time - dependence and dose - dependence. The injury is obviously improved after giving galuteotin and flavonoids from Galium verum L.
