高效液相色谱法同时测定吴茱萸汤中5种活性成分的含量

摘　　要：目的建立吴茱萸汤中吴茱萸碱、吴茱萸内酯、吴茱萸次碱、6-姜酚、芦丁5种活性成分含量同时测定的方法。方法采用HPLC法,色谱柱:COSMOSIL 5 C18-MS-II(5μm,4.6 mm×250 mm);流动相:乙腈(0.1%磷酸A)-水溶液(0.1%磷酸B),梯度洗脱,流速1 m L·min-1,柱温30℃,检测波长210 nm。结果吴茱萸碱、吴茱萸内酯、吴茱萸次碱、6-姜酚、芦丁分别在0.0098~0.196、0.205~4.10、0.0026~0.052、0.039~0.78、0.017~0.34μg内与峰面积呈良好的线性关系,r分别为0.9998、0.9999、0.9997、0.9996、0.9998,加样回收率分别为95.2%、98.8%、96.4%、99.9%、95.2%。结论该方法快速简便,重复性好,专属性强,可作为吴茱萸汤的质量控制标准和方法。Objective To simultneously determine the contents of 5 ingredients in Wuzhuyu decoction by HPLC method. Methods COSMOSIL 5 C18-MS-Ⅱ （5 μm, 4.6 mm × 250 mm） column with the mobile phase of acetonitrile and 0.1% acetic acid aqueous solution in gradient elution mode was used. The flow rate was 1.0 mL . min-1 at 30 ℃, and the injection volume was 10 μL. The detection wavelength was 210 nm. Results Evodiamine, evodine, rutecarpine, 6-gingerol and rutin had good linearity at 0.0098 - 0.196, 0.205 - 4.10, 0.0026 - 0.052, 0.039 - 0.78, and 0.017 - 0.34 μg, respectively （r = 0.9998, 0.9999, 0.9997, 0.9996, and 0.9998）. The recovery for evodiamine, evodine, rutecarpine, 6-gingerol and rutin was 95.2%, 98.8%, 96.4%, 99.9% and 95.2%. Conclusion The method is simple, precise, accurate and specific for the determination of 5 constituents in Wu- zhuyu decoction, and can be used for the quality control of Wuzhuyu decoction.
