摘 要:目的:探讨芪桂益脉灵(QGYML)对急性心肌缺血再灌注损伤大鼠心肌组织PI-3Kp85a(磷酸肌醇-3激酶)、AKT2(丝氨酸-苏氨酸激酶)mRNA的影响。方法:取SD大鼠50只,随机分为5组,正常组、假手术组、模型组、芪桂益脉灵低剂量组和芪桂益脉灵高剂量组,分别给予生理盐水、QGYML 0.6、1.2 g/kg灌胃,每天1次,连续灌胃7 d。处死大鼠,取心尖部缺血组织,采用半定量逆转录-聚合酶链反应(RT-PCR)方法测定PI-3Kp85a mRNA、AKT2 mRNA的表达。结果:与正常组、假手术组比较,模型组大鼠心肌组织PI-3Kp85a mRNA、AKT2 mRNA表达明显减弱,芪桂益脉灵组PI-3Kp85a mRNA、AKT2m RNA表达明显增强,与模型组比较具有统计学意义(P<0.05),高剂量组的效果更加显著。结论:芪桂益脉灵可显著提高大鼠缺血再灌注损伤心肌组织PI-3Kp85a mRNA、AKT2 mRNA的表达水平,对心肌具有保护作用。Objective: To explore the effects of Qigui Yimailing(QGYML) on PI -3Kp85a mRNA and AKT2 mRNA expressions in myocardial tissue of rat with ischemia reperfusion injury. Methods:50 SD male rats were randomly divided into 5 groups ,each had 10 rats. The normal group(N) after 7 days physiological saline ir- rigation didn't do any processing, the sham operation group (SH) after 7 days physiological saline irrigation took the thoracotomy to expose left coronary artery of the rats ,thread but did not make ligation, the rest rats were di- vided into pure ischemia reperfusion group , QGYML low dose group(QL) and QGYML high dose group( QH), with the physiological saline,QGYML( 1.2 g/kg and 0. 6 g/kg) respectively,whose irrigation were served 7 days in a row during the morning. Acute myocardial infarction(AMI) models were caused by ligation of the left anteri- or descending branch of the coronary artery in the successful rat models' hearts. After execution of rats, RT - PCR was used to determine the expressions of PI -3Kp85a mRNA and AKT2 mRNA in the tip of the rat cardiac is- chemic tissue. Results:PI -3Kp85a mRNA and AKT2 mRNA expressions of model group were significantly re- duced, the differences were statistic significant to compare with normal or sham group( P 〈 0. 05 ) ;PI -3Kp85a mRNA and AKT2 mRNA expressions of QGYML groups were obviously higher than those of model group (P 〈 0. 05 ), and the effect of QGYML was more significant. Conclusion: QGYML can improve the expressions of PI - 3Kp85a mRNA and AKT2 mRNA in the myocardial tissue of rats with ischemia -reperfusion injury, which can protect damaged myocardium.
