摘 要:目的探讨中药红景天苷对高糖培养的血管平滑肌细胞(VSMCs)增殖和线粒体动力学的调控作用及机制。方法将对数增殖期的VSMCs随机分为低糖(5mmol/L)培养和高糖(25mmol/L)培养两组,每组再分为5个亚组,分别在普通培养基(对照组)、低浓度红景天苷(0.3mmol/L)、高浓度红景天苷(0.5mmol/L)、低浓度Mdivi-1(10μmol/L)、高浓度Mdivi-1(25μmol/L)5种干预条件下培养24h,采用Western Blot和real time PCR检测线粒体动力学相关蛋白Drp1和Mfn2的表达水平,采用激光扫描共聚焦显微镜观察线粒体内膜情况,Image J软件分析线粒体碎片计数(MFC)。后用siRNA抑制Mfn2,观察VSMCs增值情况。结果在高糖培养条件下,VSMCs过度增殖,同时线粒体分裂增多,Drp1表达增加,Mfn2表达减少。红景天苷和Mdivi-1干预条件下,VSMCs,线粒体分裂,Drp1表达均明显受抑制,而Mfn2表达增加。抑制Mfn2表达,VSMCs增殖部分恢复高糖水平。结论红景天苷可以部分通过抑制线粒体分裂(下调Drp1和上调Mfn2)抑制高糖诱导的血管平滑肌细胞过度增殖。Objective To investigate the impact of salidroside on vascular smooth muscle cells(VSMCs)proliferation and mitochondrial dynamics under high glucose conditions,and its possible mechanism.Methods The VSMCs in proliferative phase were randomly divided into two groups:low glucose group cultured low glucose(5 mmol/L),and high glucose group cultured high glucose(25mmol/L).Then VSMCs in each group were divided into five subgroups:control group,low concentration salidroside group(0.3 mmol/L),high concentration salidroside group(0.5 mmol/L),low concentration Mdivi-1 group(10μmol/L),high concentration Mdivi-1 group(25μmol/L).VSMCs were cultured by low or high concentration of salidroside or Mdivi-1 under high glucose or low glucose condition for 24 hours.The expressions of proteins related to mitochondrial dynamics including dynamin-related protein(Drp1)and mitofusin 2(Mfn2)were tested by western blot and real-time polymerase chain reaction(PCR).The mitochondria inner membrane was observed under laser scanning confocal microscope(LSCM).The mitochondrial fission count(MFC)was analyzed by Image J software.Then Mfn2 was inhibited by small interfering RNA(siRNA).VSMCs proliferation was observed.Results VSMCs was abnormally proliferated,mitochondrial fission increased,Drp1 expression increased,and Mfn2 expression reduced under high glucose condition.Salidroside and an inhibitor of Drp1(Mdivi-1)can inhibit VSMCs proliferation,Drp1 expression and mitochondrial fission and up-regulate Mfn2 expression(all P〈0.05).After using siRNA to silence Mfn2 expression,the inhibitive effect on VSMCs proliferation can be partly reversed.Conclusion Salidroside can inhibit VSMCs proliferation induced by high-glucose and may act its therapeutic effect via inhibiting mitochondrial fission by down-regulation of Drp1 expression and up-regulation of Mfn2 expression.
