摘  要:目的观察银杏提取物EGb761对小鼠肺腺癌血管生成的影响。方法构建小鼠肺腺癌模型,用银杏提取物EGb761腹腔注射给药,在10d、20d、30d、40d观察小鼠的整体状态,计算肺组织肿瘤转移结节数目、瘤重及抑瘤率,检测肿瘤组织中血管内皮生长因子(VEGF)、肿瘤血管生长(MVD)、凝血酶敏感蛋白-1(TSP-1)的表达。结果 EGb761各剂量组的瘤重及肺转移灶数目均低于模型组,EGb761各剂量组VEGF表达、肿瘤血管密度均低于模型组,EGb761各组TSP-1显著增高,均具有显著性差异(P<0.05)。结论在小鼠肺腺癌模型中,银杏提取物EGb761可抑制小鼠肺腺癌肿瘤血管生成,其机制可能是通过调节VEGF表达减少,TSP-1表达增高而达到的。Objective To observe EGb761 effects on lung adenocarcinoma angiogenesis in mice. Methods To build the mice model of lung adenocarcinoma,administered by intraperitoneal injection of EGb761,in 10 day,20 day,30 day,40 day to observe the general condition of mice,calculation of tumor weight,inhibitory rate and lung metastatic nodules,detection expression of tumor tissue of vascular endothelial growth factor(VEGF) and tumor blood vessel growth(MVD),thrombin sensitive protein-1(TSP-1).Results Tumor weight and number of pulmonary metastases each dose group of EGb761 were lower than that of model group,VEGF expression and tumor vascular density of each dose group of EGb761 were lower than the model group. EGb761 groups significantly higher TSP-1,have significant difference(P0.05). Conclusion In lung adenocarcinoma in mice model,EGb761 can significantly inhibit the growth of lung adenocarcinoma,its mechanism may be through reducing the expression of VEGF,improve the expression of TSP-1 to inhibit angiogenesis and tumor nutrition supply,thus affecting tumor growth.
