两种人NK细胞培养体系相关性研究

摘  要:目的探讨人自然杀伤(NK)细胞小规模研究型培养体系与大规模临床制备型培养体系的相关性。方法分离11例健康志愿者的外周血,制备外周血单个核细胞(PBMCs),分别于小规模研究型培养体系与大规模临床制备型培养体系中进行诱导和扩增。比较两种培养体系下,人NK细胞的增殖倍数、细胞表型、相关活化受体以及细胞毒作用。结果第14天,两种培养体系下人NK细胞的增殖倍数、细胞表型以及细胞毒作用的方面比较,差异均无统计学意义(P>0.05)。结论人NK细胞小规模研究型培养体系和临床制备型培养体系具有较好的一致性,前者可为后者的建立与优化提供稳定的研究模型,且能明显降低研究成本。Objective To explore the comparative relationship between small-scale research culture system and clinical grade preparative culture system in generating of human natural killer( NK) cells. Methods Peripheral blood mononuclear cells( PBMCs) isolated from 11 healthy volunteers were induced and amplified in the above two NK cells culture systems in vitro. Multiples of NK cell proliferation,cell phenotype,activating receptor and cytotoxicity were analyzed. Results At the end of culture( day 14),there were no significant differences in cell proliferation,cell phenotype and cytotoxicity between the two NK cell culture systems( P 0. 05).Conclusion Small-scale research culture system can be used as a more cost-effective stable research model for establishment and optimization of human NK cell production in lieu of a clinical grade preparative culture system.
