摘 要:目的观察仁术健脾理气颗粒对功能性消化不良(FD)脾虚气滞证大鼠胃排空及胃肠激素的影响。方法 SD大鼠80只,随机分为正常组20只和造模组60只。造模组采用大承气汤灌胃+饮食失调法+郭氏夹尾应激法,诱导FD脾虚气滞证大鼠模型。造模第10天,分别从正常组和造模组各随机抽取10只大鼠进行造模成功评定。将造模组剩余的50只大鼠随机分为模型组,仁术健脾理气颗粒高、中、低剂量组,吗丁啉组,每组10只,分别给予生理盐水或药物干预3周后,检测各组大鼠体重变化、易激怒程度评分、3 h进食量及胃排空率,采用酶联免疫分析法检测大鼠血清生长抑素(SS)、胆囊收缩素(CCK)及胃泌素(GAS)水平。结果仁术健脾理气颗粒各剂量组大鼠胃排空率均高于模型组(P<0.05,P<0.01),血清SS、CCK水平均低于模型组(P<0.01,P<0.05),血清GAS水平高于模型组(P<0.01)。结论仁术健脾理气颗粒可促进FD脾虚气滞证大鼠胃排空功能,调节胃肠激素水平。Objective To observe the influence of Renzhu Jianpi Liqi Granules( RG) on gastric emptying and gastrointestinal hormone in rats of spleen deficiency and qi stagnation syndrome with functional dyspepsia( FD). Methods 80 SD rats were randomly divided into the normal group( 20) and model-building group( 60). The model-building group adopted Dachengqi Decoction for gavage and diet imbalance method combined with the clip tail stimulation stress method in order to induce FD model rats with spleen deficiency and qi stagnation syndrome. On the 10 th days,10 rats were randomly chosen from both two groups for model success evaluation. The remaining 50 rats of model group were randomly divided into the model group with RG in high,medium and low dose and Domperidone group( 10 each),and were given normal saline or drug intervention respectively for 3 weeks,then the change of weight,the scores of being easy to provoke,food intake of 3 hours and gastric emptying rate were tested in each group, ELISA was used to detect serum somatostatin( SS),cholecystokinin( CCK) and gastrin( GAS) levels. Results The gastric emptying rate of each dose RG groups were significantly higher than that of the model group( P 〈 0. 05,P〈 0. 01); serum SS,CCK levels were lower than that of the model group( P 〈0. 05,P 〈 0. 01); serum GAS were significantly higher than that of the model group( P 〈 0. 01). Conclusion RG can accelerate gastric emptying function in FD rats with spleen deficient and qi stagnation syndrome and adjust gastrointestinal hormone levels.
