通窍活血汤对血管性痴呆大鼠cAMP和PKA表达的调控作用

摘　　要：目的:探讨通窍活血汤对血管性痴呆大鼠血清中环磷酸腺苷(c AMP)、蛋白激酶A(PKA)表达的影响。方法:将50只大鼠按照随机数字表法分为假手术组、模型组及通窍活血汤高、中、低剂量组,间断夹闭颈总动脉法建立血管性痴呆模型,用Morris水迷宫实验检测模型是否成功,利用酶联免疫分析(ELISA)检测血清中c AMP、PKA的含量。结果:与假手术组比较,模型组大鼠血清中c AMP、PKA的含量均降低(P<0.01);通窍活血汤高、中剂量组血清中c AMP、PKA的含量较模型组升高(P<0.01)。结论:通窍活血汤可增加血管性痴呆大鼠血清中c AMP、PKA的含量,增强c AMP-PKA表达的作用。Objective：To investigate the effects of Tongqiao Huoxue decoction on expression levels of cyclic adenosine monophosphate（c AMP） and protein kinase A（PKA） in rats with vascular dementia（VD）. Methods：The rats were randomly divided into sham operation group,VD model group,the large,middle and small dose Tongqiao Huoxue treatment groups. The rat model of VD was built by intermittently clipping common carotid artery. Morris water maze test determined whether the surgery had been successful. After that rats were perfused with Tongqiao Huoxue decoction,and the changes of c AMP and PKA were detected by Enzyme-linked immunosorbent assay（ELISA）. Results：The content of c AMP and PKA in VD rats were lower than those in sham operated rats（P〈0.01）. The above indexes of the large- and middle-dose Tongqiao Huoxue groups were higher compared with the VD model group（P〈0.01）. Conclusion：Tongqiao Huoxue decoction increases the content of c AMP and PKA,and thus strengthens the expression of c AMP-PKA.
