摘  要:目的探讨紫檀芪对缺氧/复氧诱导的乳鼠心肌细胞凋亡的影响,以及对磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)信号通路的影响。方法乳鼠心肌细胞经过缺氧/复氧(H/R)处理,再模拟心肌缺血再灌注损伤。实验分为正常组,模型组(H/R),紫檀芪组(H/R+0.5,5,50μmol/L紫檀芪)。MTT法检测各组心肌细胞活力;倒置显微镜拍照检测各组心肌细胞形态;Annexin V-FITC/PI流式双染法检测各组心肌细胞凋亡;Western blot分析PI3K/AKT激活状况。结果与正常组比较,模型组中心肌细胞皱缩,变圆变亮,细胞活力降低,细胞早期凋亡和晚期凋亡率显著提高,PI3K表达量及AKT磷酸化程度降低,差异均具有显著性(P<0.05);与模型组比较,5,50μmol/L紫檀芪组中心肌形态恢复,细胞活力上升,PI3K表达量提高,差异均具有统计学意义(P<0.05);0.5,5,50μmol/L紫檀芪组心肌细胞凋亡程度下降,AKT磷酸化水平上升,差异均具有统计学意义(P<0.05)。结论紫檀芪可抵抗缺氧/复氧诱导的乳鼠心肌细胞凋亡,PI3K/AKT信号被激活是其作用表达的主要方式。Objective To investigate the effect of Pterocarpus on apoptosis of neonatal rat cardiomyocytes induced by hypoxia / reoxygenation and the effect of phosphatidylinositol 3-kinase(PI3K) / protein kinase B(AKT) signaling pathway. Methods Cardiomyocytes of neonatal rats were subjected to hypoxia / reoxygenation(H / R), and the hearts were subjected to myocardial ischemia-reperfusion injury. The rats were divided into normal group, model group(H / R) and pterocarpus stilbene group(H/R+0.5,5,50μmol/L).Cell viability was measured by MTT assay. Cardiomyocyte morphology was detected by inverted microscope. Annexin V-FITC / PI flow cytometry was used to detect the apoptosis of cardiomyocytes.PI3K/AKT activation was analyzed by Western blot. Results Compared with the normal group, the center of model group of muscle cell was shrinkage, rounding and brightening, and the cell viability decreased, early and late apoptosis rate of cells increased significantly. The expression of PI3 K and the phosphorylation of AKT in the model group were significantly higher than those in the model group(P〈0.05). Compared with the model group, 5, 50mol/L of pterostilbene group center muscle morphological was recovery, cell viability was increased, and the rosewood the expression of PI3 K was increased, and the difference was significant(P〈0.05). 0.5,5,50 mol/L pterostilbene group the apoptosis of myocardial cells decreased, the phosphorylation level of AKT increased, the difference was significant(P〈0.05). Conclusion Pterocarpus can resist cardiomyocyte apoptosis induced by hypoxia / reoxygenation, and activation of PI3 K / AKT signal is the main way of its expression.
