摘  要:目的:观察脑梗死后调控血管发生通路上关键因子APJ及其配体Apelin的变化规律及针刺对其干预效应规律。方法:Wistar大鼠随机分为模型组(90只),电针组(90只),假手术组(90只)和空白组(10只),前3组又分为大脑中动脉阻滞(MCAO)后1、3、6、9、12、24h,3、7、12d共9个时相组,每组10只。各电针组电针"水沟"穴20min,1、3、6、9、12、24h组造模后即刻治疗1次,并于相应时点取材,3、7、12d组每日治疗1次,末次治疗结束后取材。应用实时荧光定量法(RT-PCR)、蛋白免疫印迹法(Western blot)检测脑血管内皮细胞Apelin、APJ mRNA及蛋白表达的变化。结果:与空白组相比,假手术组Apelin和APJ mRNA表达量差异无统计学意义(P>0.05)。与假手术组比较,模型组12h、12dApelin和APJ mRNA表达下调(P<0.05,P<0.01)。与同时相模型组比较,电针组Apelin mRNA在12h、7d的表达量上调(P<0.01);电针组APJ mRNA在6、9、12h的表达量上调(P<0.05,P<0.01)。与空白组相比,假手术组Apelin和APJ蛋白表达量差异无统计学意义(P>0.05)。与假手术组比较,模型组Apelin蛋白表达量1、3、6、24h及3、7、12d下调(P<0.01,P<0.05),模型组APJ蛋白表达量1、3、6、9h及3d下调(P<0.01,P<0.05)。与同时相模型组比较,电针组Apelin蛋白表达在6、24h及3、7、12d均上调(P<0.05,P<0.01),电针组APJ蛋白表达量于1、9、12、24h及3、7、12d上调(P<0.05,P<0.01)。结论:电针可上调MCAO大鼠脑血管内皮Apelin-APJ mRNA及蛋白的表达,这对于脑缺血后血管新生及侧支循环的建立有重要作用。Objective To observe the regulation of APJ and its ligand Apelin on the angiogenesis pathway after cerebral infarction and the intervention effect of acupuncture.Methods Wistar rats were randomly divided into model group(n=90),electroacupuncture(EA)group(n=90),sham operation group(n=90)and control group(n=10).The first three groups were further divided into 1,3,6,9,12,24 hand 3,7,12dsubgroups(n=10in each subgroup).The cerebral infarction model was established by middle cerebral artery occlusion(MCAO).EA(15Hz,2mA)was applied to"Shuigou"(GV 26)for 20 min in the EA group.The 1,3,6,9,12,24 hsubgroups were treated immediately after modeling,the 3,7,9dsubgroups were treated once daily for 3,7or 9days.Real-time fluorescent quantitative(RT-PCR)and Western blot were applied to detect the changes of Apelin and APJ in cerebrovascular endothelial cells,respectively.Results Compared with the control group,the expression of Apelin-APJ mRNA was decreased in the model group(12h,12 d,P0.05,P0.01);After EA,the Apelin mRNA expression was increased in the 12 hand 7dsubgroups(P0.01),while the APJ mRNA expression was increased in the 6,9,12hsubgroups(P0.05,P0.01).Compared with the control group,the Apelin(1,3,6,24 hand 3,7,12d)and APJ(1,3,6,9hand 3d)protein expressions were decreased in the model group(P0.01,P0.05);After EA,the Apelin protein expression was increased in the 6,24 hand 3,7,12dsubgroups(P0.05,P0.01),while the APJ protein expression was increased in the 1,9,12,24 hand 3,7,12dsubgroups(P0.05,P0.01).Conclusion EA can up-regulate the expression of Apelin-APJ mRNA and protein of cerebral vascular endothelial cell in MCAO rats which has an important role in the establishment of blood vessel regeneration and collateral circulation.
