摘 要:目的:探讨双强度、频度参数交叉配对电针对血管性痴呆(VD)大鼠海马CA 1区β淀粉样蛋白1-40(Aβ1-40)、精氨酸加压素(AVP)及学习记忆的影响。方法:SD大鼠随机抽取8只为假手术组,余大鼠采用4-VO制造血管性痴呆动物模型,造模后筛选出合格的大鼠分为模型组、电针1组(强度0.5mA,每5d治疗1次)、电针2组(强度1.5mA,每5d治疗1次)、电针3组(强度0.5mA,每天治疗1次)、电针4组(强度1.5mA,每天治疗1次),每组8只。电针组针刺"百会""大椎"穴,共治疗10次。运用荧光定量PCR法检测大鼠海马CA 1区Aβ1-40、AVP基因表达,Morris水迷宫测试各组大鼠学习记忆能力。结果:模型组与假手术组比较,平均逃避潜伏期与首次跨越平台时间均延长,2min内跨越平台次数减少,Aβ1-40基因相对表达量增加,AVP基因相对表达量减少(P<0.05)。各电针组与模型组比较,平均逃避潜伏期与首次跨越平台时间均缩短,2min内跨越平台次数增多,Aβ1-40基因相对表达量减少,AVP基因相对表达量增加(P<0.05)。各电针组间进行比较,同频度不同强度时,高强度组(电针2组、电针4组)较低强度组(电针1组、电针3组)大鼠的学习记忆能力强,海马CA 1区Aβ1-40基因相对表达量减少、AVP基因相对表达量增加(P<0.05);同强度不同频度时,高频度组(电针3组、电针4组)较低频度组(电针1组、电针2组)大鼠的学习记忆能力强,海马CA 1区Aβ1-40基因相对表达量减少、AVP基因相对表达量增加(P<0.05)。结论:电针可有效地改善VD大鼠学习记忆能力,其机制可能为电针抑制海马CA 1区Aβ1-40、促进AVP的基因表达;针效高强度优于低强度,高频度优于低频度,高强度与高频度配对治疗方案最优。Objective To observe the influence of electroacupuncture(EA)stimulation with different electrical current intensities and therapeutic intervals on learning-memory ability and expression ofβ-amyloid peptide Aβ1-40 and arginine vasopressin(AVP)genes in the hippocampal CA 1region in vascular dementia(VD)rats,so as to provide evidence for treatment of VD.Methods A total of 48 male SD rats were randomly divided into sham,model,0.5mA-5d-EA,1.5mA-5d-EA,0.5mA-1d-EA and 1.5 mA-1d-EA groups(n=8in each group).The VD model was established using modified 4-vessels occlusion method.EA(alternative 2Hz/15 Hz,0.5mA,1.5mA)was applied to"Baihui"(GV 20)and"Dazhui"(GV 14)for 30 min,once a day(1d)or once every 5dfor 10 times.The learning-memory ability was detected using Morris water maze tests(place navigation task and spatial probe trials),and the expression of Aβ1-40 and AVP genes in hippocampal CA 1region was determined using real time-PCR.Results In comparison with the sham group,the average escape latency of place navigation task,and the duration for crossing the target-platform for the 1st time(spatial navigation task)were significantly increased(P0.05),and the times to cross the target-platform within 2min(spatial probe trials)were significantly decreased in the model group(P0.05),suggesting a reduction of learning-memory ability.The expression level of Aβ1-40 mRNA was remarkably increased(P0.05),and that of AVP mRNA notably decreased in the model group(P0.05).Following EA intervention,the increased escape latency and the duration of crossing the target-platform for the 1st time and the increased Aβ1-40 mRNA expression,and the decreased target-platform crossing times within 2min as well the down-regulated AVP mRNA expression level were reversed in the 4EA groups compared with the model group(P0.05).The therapeutic effects of higher stimulating intensity(1.5mA-1d-EA and1.5mA-5d-EA)were markedly superior to those of lower intensity(0.5mA-1d-EA and
