摘 要:以改良MS为基本培养基,研究青钱柳不同取样时间、取样部位、低温处理、外植体接种方式及不同植物生长调节剂对其茎段初代培养的影响。结果表明,青钱柳茎段培养最佳取样时间为5月,顶芽下1、2段萌芽率最高;4℃低温处理4 h及外植体竖放能明显降低初代培养褐化率,提高萌芽率;青钱柳茎段初代培养最适宜培养基为改良MS+TDZ 0.5 mg/L+NAA 0.05 mg/L。In this study, different plant growth re the effects of sampling time, sampling position, low temperature, inoculation types of explants and gulators on primary culture of stem segments of Cyclocarya paliurus were studied on improved MS medium. The results showed that May was the best time for collecting explant. The first and the second nodes from the top had highest germination rate as explants. The browning rate reduced obviously when the explants were been treated by 4℃ for 4 hours and placed upright, while the germination rate increased. The optimal hormone combination was TDZ 0.5mg/ L+NAA 0.05mg/L.
