摘  要:报道了一类新型的以锍离子提供正电荷的阳离子脂质体的合成。以四氢噻吩和四氢噻喃为原料,在四氟硼酸银体系中与链状碘代脂肪烷烃经取代反应制得硫正离子化合物,产物用氯离子交换树脂交换得阴离子为氯离子的8种新型的链状锍类鎓氯盐脂质体化合物,其结构经~1H NMR,^(13)C NMR,FT-IR和HR-MS(ESI)表征。凝胶电泳阻滞实验表明该类锍化合物具有和DNA聚合并形成复合物的能力,随着脂肪链的增长,复合能力增强。A new type of cationic lipid composed with sulfonium as the positive charge was reported.The sulfonium compounds were synthesized starting from tetrahedrothiophene and tetrahedrothiopyran by reaction with lipid iodides in the presence of silver tetrafluoro borate.The products were treated with chloro type anion exchange resin to give eight novel sulfonium chloride compounds.The structures were characterized by ^1H NMR,^13C NMR,FT-IR and HR-MS(ESI).Gel retardation experiments showed that the sulfonium compounds can condense DNA and form the lipopolyplex effectively.The results revealed that compound with longer lipid chain have stronger condensing effect.
