摘 要:目的观察女贞子抗CCl_4致小鼠肝损伤的作用,并探讨其可能的作用机制。方法将昆明种小鼠随机分为模型组、女贞子水提物组和齐墩果酸组,分别预防性给予各组小鼠相应药物4 d后,ip给予各组小鼠CCl_4造模,处死小鼠后,计算肝脏指数和测定血清丙氨酸转氨酶(ALT)的含量;用实时荧光定量PCR法检测肝脏早期生长反应基因1(EGR-1)、巨噬细胞炎症蛋白2(MIP-2)和角化细胞源性趋化因子(KC)mRNA的表达。结果与模型组比较,女贞子水提物能显著降低小鼠血清中ALT的含量,使肝脏炎症反应基因EGR-1、MIP-2和KC mRNA的表达显著降低。结论女贞子抗CCl_4致小鼠肝损伤的作用机制可能与抑制肝脏炎症反应基因的表达有关。OBJECTIVE To observe the against hepatic injury effect in mice of Ligustri lucidifructus (LLF) induced by CCl4, and to explore its possible mechanism. METHODS Kunming mice were randomly divided into model group, water extracts group of LLF and oleanolic acid group. The mice in each group were administered with corresponding drugs. After 4 days,the CCl4 liver injury model was established by intraperitoneal injection, and all mice were killed. The liver index and serum alanine aminotransferase (ALT) content were calculated, and the mRNA expressions of liver early growth response gene - 1 ( EGR - 1 ) , macrophage inflammatory protein - 2 ( MIP - 2 ) and keratinocyte derived chemokine ( KC ) were detected by real - time RT - PCR. RESULTS Compared with the model group, the water extracts of LLF could significantly reduce the content of ALT in serum, and the hepatic inflammatory response genes of Egr - 1, MIP - 2 and KC mRNA were decreased significantly. CONCLUSION The mechanism of LLF against hepatic damage induced by CC14 may be related to the inhibition of the expression of hepatic inflammatory response genes.
