摘  要:目的探讨红花注射液对盆腔炎模型大鼠子宫组织中左氧氟沙星分布的影响。方法将96只盆腔炎模型大鼠按照完全随机分组法分为实验组(左氧氟沙星和红花注射液联合用药组)和对照组(单用左氧氟沙星组),每组48只,实验组动物连续3 d静注红花注射液(0.8 m L/kg),对照组肌注等容积生理盐水。第4天,两组动物均按80 mg/kg灌服左氧氟沙星,于给药后0.25、0.5、1、1.5、2、4、8、12 h两组分别处死大鼠6只,采集动物血浆,并取子宫组织制作匀浆,用高效液相色谱法测定血浆和子宫组织匀浆中左氧氟沙星浓度,DAS 2软件计算药动学参数,如达峰浓度(Cmax)、达峰时间(tmax)、半衰期(t1/2β)和药-时曲线下面积(AUC0→t)等。结果实验组和对照组血浆及子宫组织的药-时曲线均符合二室模型,其药动学参数如下:血浆Cmax分别为(14.28±0.84)、(14.45±1.93)μg/m L,子宫组织Cmax分别为(15.85±1.32)、(12.65±1.01)μg/m L(P<0.01);血浆药物清除率分别为(1.25±0.18)、(1.35±0.25)L/(kg·h),子宫组织药物清除率分别为(1.08±0.08)、(1.47±0.16)L/(kg·h)(P<0.01);血浆tmax分别为(1.42±0.20)、(1.50±0.01)h,子宫组织tmax分别为(1.25±0.27)、(1.17±0.26)h;血浆t1/2β分别为(7.69±2.61)、(7.32±2.80)h,子宫组织t1/2β分别为(7.95±5.29)、(9.16±3.00)h;血浆AUC0→t分别为(55.13±5.42)、(51.73±5.27)μg·h/m L,子宫组织AUC0→t分别为(63.69±3.95)、(48.95±4.11)μg·h/m L(P<0.01)。结论红花注射液可以明显提高左氧氟沙星在盆腔炎模型大鼠子宫组织中的药物浓度。Objective To study the effect of Honghua Injection on the distribution of levofloxacin( LVFX) in the uterine tissue of pelvic inflammatory rat model. Methods Ninety-six female rats with bacterial pelvic inflammation were randomly divided into( n = 48 per group) experimental group( combined treatment of Honghua and LVFX) and control group( only LVFX). Rats in experimental group were intramuscularly injected with Honghua Injection( 0. 8 m L / kg) for 3 consecutive days,and those in the control group were given the same volume of normal saline. On the day 4,all the rats were orally administered with LVFX( 80 mg / kg). Six rats of each group were sacrificed at each time point of 0. 25,0. 5,1,1. 5,2,4,8and 12 h after Honghua Injection for collecting the plasma and uterine samples. Concentrations of LVFX in the plasma and uterine homogenous tissues were detected by high performance liquid chromatography( HPLC),and pharmacokinetic parameters were calculated with DAS 2 software. Results Concentration-time curves of LVFX of the both groups fit the two-compartment model. The values of main pharmacokinetic parameters in plasma and uterine tissue of experiment and control groups were listed as follows: Cmaxwere 14. 28 ± 0. 84 and14. 45 ± 1. 93 μg / mL for plasma,15. 85 ± 1. 32 and 12. 65 ± 1. 01 μg / mL for uterine; tmaxwere 1. 42 ± 0. 20 and 1. 50 ± 0. 01 h for plasma,1. 25 ± 0. 27 and 1. 17 ± 0. 26 h for uterine; t1 /2βwere 7. 69 ± 2. 61 and 7. 32 ±2. 80 h for plasma,7. 95 ± 5. 29 and 9. 16 ± 3. 00 h for uterine; Clearance( CL) were 1. 25 ± 0. 18 and1. 35 ± 0. 25 L /( kg·h) for plasma,1. 08 ± 0. 08 and 1. 47 ± 0. 16 L /( kg·h) for uterine; Area under curve( AUC)0→twere 55. 13 ± 5. 42 and 51. 73 ± 5. 27 μg · h / mL for plasma,63. 69 ± 3. 95 and 48. 95 ± 4. 11μg·h / mL for uterine. Moreover,the differences of parameters Cmax,CL and AUC0→tin the uterine tissue were statistically significant between the 2 groups( P〈0. 01). Conclusion Honghua I
