摘  要:采用HPLC法测定菊芋叶和向日葵叶中绿原酸含量,并对收获期菊芋叶和向日葵叶中绿原酸的积累分布进行研究。色谱条件为:ZORBAX Extend-C_(18)(4.6mm×250mm,5μm),柱温30℃,检测波长327nm,进样量10μL,流速1.0mL·min^(-1),流动相为乙腈-水(含体积分数为0.05%的甲酸),梯度洗脱。提取条件:甲醇-水(70∶30,体积比)回流浸提,料液比1∶25(g∶mL),提取时间1.5h,提取温度60℃。结果表明,菊芋叶和向日葵叶中的绿原酸在收获期积累分布差异显著,菊芋叶中绿原酸含量在10月下旬最高,为4.20%;向日葵叶中绿原酸含量在8月下旬最高,为0.47%。因此,10月下旬的菊芋叶可以作为提取绿原酸的植物原材料。The contents of chlorogenic acid(3-CQA)in Helianthus tuberosus L.leaves and Helianthusann uusL.leaves were determinated by HPLC.The content accumulated distributions of 3-CQA in Helianthus tuberosus L.leaves and Helianthus annuus L.leaves at harvest time were investigated.The chromatographic conditions were as follows:a ZORBAX Extend-C18column(4.6 mm×250 mm,5μm),column temperature of 30 ℃,detection wavelength of 327 nm,injection volume of 10μL,flow rate of 1.0mL·min-1,gradient mobile phase of acetonitrile-water(containing 0.05%formic acid).The optimal extraction conditions were as follows:solvent refluxing extraction with methanol-water(70∶30,volume fraction),material-liquid ratio of 1∶25(g∶mL),extraction time of 1.5h,extraction temperature of 60℃.The results demonstrated that the content accumulated distributions of 3-CQA in Helianthus tuberosus L.leaves and HelianthusannuusL.leaves were significant different at harvest time.The content of 3-CQA in Helianthus tuberosus L.leaves reached the highest level(4.20%)in late October.The content of 3-CQA in Helianthus annuus L.leaves reached the highest level(0.47%)in late August.Thus,Helianthus tuberosus L.leaves in late October could be used as the raw material for extracting chlorogenic acid.
