摘 要:目的研究人胎盘间充质干细胞(hpMSCs)对呼吸道合胞病毒(RSV)致毛细支气管炎大鼠肺组织中视黄酸受体相关孤核受体γt(RORγt)的调控及其意义。方法将36只sD幼鼠随机分为正常对照组、RSV致毛细支气管炎组(RSV组)和hpMSCs组,每组12只。RSV组和hpMSCs组均采用RSV滴鼻法建立毛细支气管炎动物模型,hpMSCs组接种病毒后向大鼠尾静脉内注射hpMSCs进行干细胞移植治疗,正常对照组不制作模型。观察3组肺组织病理学变化;应用酶联免疫吸附试验检测血浆白细胞介素17(IL-17)、IL-23的含量;采用实时荧光定量聚合酶链反应检测各组外周血单个核细胞(PBMC)及肺组织中RORγtmRNA的水平;采用蛋白质印迹法检测3组肺组织中RORγt蛋白的表达。结果RSV组大鼠肺组织肺间质炎症明显,肺泡间隔增宽,肺泡腔变小,有大量嗜酸粒细胞、淋巴细胞浸润;与RSV组相比,hpMSCs组肺间质炎症明显减轻,肺泡腔相对增大,炎性细胞浸润减少;正常对照组大鼠肺组织无炎性改变。RSV组血浆IL-17、IL-23含量高于正常对照组[(56±6)ng/L比(27±4)ng/L、(61±5)ng/L比(19±3)ng/L](P〈0.05);hpMSCs组IL-17、IL-23含量[(30±4)ng/L、(40±6)ng/L]低于RSV组(P〈0.05)。RSV组大鼠PBMC及肺组织中RORTγt mRNA表达水平均高于正常对照组(PBMC:11.4±1.1比1.0±0.0,肺组织:8.1±0.7比1.0±0.0)(P〈0.05);hpMSCs组PBMC及肺组织中RORγtmRNA表达水平(PBMC:4.4±0.7.肺组织:3.4±0.5)均低于RSV组(均P〈0.05)。RSV组肺组织中RORγt蛋白的表达水平高于正常对照组(1.26±0.12比0.72±0.12)(P〈0.05);hpMSCs组肺组织中RORγt蛋白表达水平(0.88±0.14)低于RSV组(P〈0.05)。结论hpMSCs可以下调RSV致毛细支气管炎大鼠肺组织中特异性转录因子RORγt及相关细胞因子的表达水平,�Objective To study the regulation of human placenta derived mesenchymal stem cells (hpMSCs) and retinoid-related orphan receptor γt (RORγt) in lung tissue of capillary bronchitis rats caused by respiratory syncytial virus (RSV). Methods A total of SD 36 juvenile rats were randomly divided into normal group, RSV group and hpMSCs group, 12 rats in each group. The RSV group and hpMSCs group had capillary bronchitis models induced by RSV; the hpMSCs group had stem cell transplantation therapy with hpMSCs injection after virus infection. The normal control group had no treatment. Lung histopathological changes were observed in 3 groups. Plasma levels of interleukin( IL)-17 and IL-23 were tested by enzyme linked immunosorbent assay. Expressions of RORγt in peripheral blood mononuclear cell (PBMC) and lung tissue were tested by real-time fluorescence quantitative polymerase chain reaction and western blotting method. Results The RSV group had lung interstitial hyperemia, edema, inflammatory cell infiltration, wide alveolar septa and bronchial collapse and deformation. The lung inflammatory manifestation in hpMSCs group was obviously ameliorated compared to that in RSV group. Levels of IL-17, IL-23 in RSV group were higher than those in normal control group [ (56 ± 6)ng/L vs (27 ±4)ng/L, (61 ±5) ng/L vs (19 ±3)ng/L] ; levels of IL-17, IL-23 in hpMSCs goup were lower than that in RSV group [ ( 30 ± 4) ng/L, (40 ± 6 ) ng/L 1 ; the differences were statistically significant ( P 〈 0. 05 ). Expressionlevels of RORγt mRNA in PBMC and lung tissue in RSV group were higher than those in normal control group (PBMC : 11.4 ± 1.1 vs 1.0 ± 0. 0, lung tissue : 8.1 ± 0. 7 vs 1.0 ± 0. 0) ; the levels of RORγt mRNA in PBMC and lung tissue in hpMSCs group ( PBMC : 4. 4 ± 0. 7, lung tissue : 3.4 ± 0. 5 ) were lower than those in RSV group; the expression level of RORγt protein in lung tissue in RSV group was higher than that in normal control group( 1. 26
