摘 要:目的:观察当归四逆汤对心肌缺血大鼠心肌SOD及MDA表达的影响。方法:采用雄性Wistar大鼠腹腔注射垂体后叶素(Pit)(30U/kg)造成急性心肌缺血损伤模型;药物干预后,观察心尖部心肌组织病理学改变,并检测超氧化物歧化酶(SOD)和丙二醛(MDA)活性。结果:光镜下显示,空白对照组心肌纤维排列整齐,心肌细胞核位于心肌纤维的中央,胞浆着色均匀;模型组心肌纤维排列轻度紊乱,呈波浪状改变,部分心肌细胞核出现溶解;当归四逆汤高、低剂量组及阳性对照组心肌纤维排列较整齐,细胞结构未见明显病理改变。模型组SOD活性明显低于空白对照组(P<0.01),MDA含量明显高于空白对照组(P<0.01);当归四逆汤高剂量组、低剂量组及阳性对照组SOD活性均明显高于模型组(P<0.05或P<0.01),MDA含量均明显低于模型组,差异均有统计学意义(P<0.05或P<0.01);当归四逆汤低剂量组SOD活性及MDA含量均高于当归四逆汤高剂量组,但差异均无统计学意义(P>0.05)。结论:当归四逆汤能提高垂体后叶素所致急性心肌缺血大鼠SOD活性,减少MDA含量,增强机体清除氧自由基的能力,抑制脂质过氧化损伤的发生,保护心肌细胞。Objective: To observe the protective effect of Danggui Sini Dectoction (DSD) on myocardial tissue of rats with experimental myocardial ischemia. Methods: Male Wistar rats by intraperitoneal injection of Pituitrin (pit) (30 U/kg) caused by acute myocardial ischemia model and observation of apical Myocardium Pathological changes, and to detect the change of superoxide dismutase (SOD) and malondialdehyde (MDA) activity. Results: Light microscope show that the blank control group, the myocardial fibers arranged in neat rows, myocardial cells in myocardial fibers of the central, color uniform cytoplasm; model group myocardial fibers arranged in mild disorder was wavy change, part of the myocardial nuclei dissolve; DSD is high, low dose group and positive control group, the myocardial fibers arranged neatly, cell structure, there are no obvious pathological change. Compared with blank control group, model group, the SOD activity decreased significantly (P〈0.01), MDA content increased significantly, and the difference is significant (P〈0.01); compared with the model group, DSD high dose group, low dose group and positive control group, the SOD activity was significantly increased (P〈0.05 or P〈0.01), MDA content decreased significantly, the difference is statistically significant (P〈0.05 or P〈0.01) and with the DSD to dose increased, SOD activity and MDA content of rat myocardial respectively, showed the trend of decrease, but there was no significant difference in SOD activity and MDA content between DSD high and low dose groups (P〉0.05). Conclusion: DGSND can protect rats from acute myocardial ischemia induced by pituitrin.
