摘 要:目的:观察补肾健骨汤对绝经后骨质疏松(PMOP)模型大鼠股骨颈骨密度及血清雌二醇(E_2)、白介素-6(IL-6)、骨钙素(BGP)影响。方法:选择4月龄SPF级未经交配健康的雌性大鼠,取12只为正常组(A组);采用去卵巢骨质疏松造模法,将造模成功的大鼠随机分为模型组(B组)、补肾健骨汤组(C组)及雷洛昔芬组(D组)各12只,假手术组(E组)12只大鼠只切开皮肤肌肉,取等同体积的脂肪组织,不去卵巢;灌胃8周后完成大鼠骨密度检测,处死取标本检测。结果:(1)骨密度:干预12周后,模型组大鼠骨密度低于假手术组(P<0.05);补肾健骨汤组及雷洛昔芬组骨密度高于模型组(P<0.05)。(2)BGP:模型组大鼠组低于假手术组(P<0.05),补肾健骨汤组及雷洛昔芬组血清BGP均高于模型组(P<0.05)。(3)雌激素:模型组雌激素水平显著低于正常组、假手术组(P<0.05);补肾健骨汤组、雷洛昔芬组雌激素水平均高于模型组,差异有统计学意义(P<0.05);雷洛昔芬组骨与补肾健骨汤组之间差异无统计学意义(P>0.05)。(4)IL-6:模型组IL-6水平明显高于正常组、假手术组,差异有统计学意义(P<0.05);补肾健骨汤组、雷洛昔芬组IL-6水平均低于模型组,差异有统计学意义(P<0.05);雷洛昔芬组骨与补肾健骨汤组之间差异无统计学意义(P>0.05)。结论:补肾健骨汤能改善去卵巢大鼠骨密度,升高雌激素及骨钙素水平,促进骨生成,并降低IL-6水平,提示这可能是补肾健骨汤防治绝经后骨质疏松症的机理之一。Objective: To observe the effect of BSJGT on Bone mineral density and the serum Estradiol (E2), lnterleukin-6 (IL-6) and bone gla protein (BGP) in the postmenopausal osteoporosis model of ovariec- tomized rat. Methods: The 60 healthy female rats, which is 4 months old and no mating, were slected and 12 rats were as a normal group (A). PMOP rats as Osteoporosis model and the Osteoporosis model rats were random divided into control group (B), BSJGT group (C), raloxifene group (D), 12 rats in each. The 12 rats of the Sham resection group (E) were only cut the skin and muscle, taken the same volume of adipose tissue, keeping the ovary. After 8 weeks of intragastric administration, the rats were subjected to bone min- eral density test. Results: (1)Bone mineral density: Compared to sham resection group, the bone mineral den- sity reduced obviously in model group, and the difference was statistically significant (P〈0.05);both of the BSJGT and raloxifene groups showed higher bone mineral density than the model group, and the difference was statistically significant (P〈0.05). (2)BGP: The model group showed lower BGP than the sham resection group (P〈0.05), and the BSJGT and raloxifene groups showed higher BGP than model group (P〈0.05). (3) E2: The model group showed lower E2 level than the control group and sham resection group (P〈0.05). The BSJGT and raloxifene groups showed higher E2 than model group (P〈0.05); there was no statistical difference between the BSJGT and raloxifene groups (P〉0.05). (4) IL-6: The model group showed higher IL-6 level than the control group and sham resection group (P〈0.05). The BSJGT and raloxifene groups showed lower IL-6 than model group (P〈0.05); there was no statistical difference between the BSJGT and raloxifene groups(P〉0.05).
