摘 要:目的:观察止带方加减不同剂量和给药方式对宫颈炎大鼠的疗效。方法:按体质量分层随机将大鼠分成6组,空白组、模型组、中药中剂量组、中药高剂量组、中药阴道给药组和双唑秦组。除空白组外,其余各组用苯酚胶浆阴道造模3 d,构建宫颈炎模型;造模后给予相应药物干预,空白组予生理盐水干预,14 d后测定大鼠体质量、受孕率、肉眼下阴道病理评分、阴道与宫颈HE染色、血清IL-6、IL-8水平。结果:与空白组比较,模型组大鼠阴道病理评分、IL-6和IL-8均显著增高(P<0.05),受孕率较低,同时阴道及宫颈可见明显病理变化;与模型组比较,药物干预的大鼠受孕率均增高,同时阴道及宫颈病变减轻;中药阴道给药组阴道病理评分、IL-6和IL-8均显著降低(P<0.05);中药中剂量组和双唑秦组IL-6、IL-8水平均显著降低(P<0.05)。结论:止带方可能是改善了局部免疫功能,降低IL-6、IL-8水平,从而减轻了宫颈炎病变程度,提高大鼠受孕率。Objective : The study aimed to explore the efficacy of Zhidai Decoction in the treatment of cervicitis rats with different doses and administration. Methods:The rats were randomly divided into the following six groups according to body weight ( ten rats in each group) : ( G1 ) normal group ( gavage saline) , ( G2 ) model ( gavage saline ) , ( G3 ) middle dose group( gavage middle - dose Zhidai Decoction), ( G4 ) high dose group( garage high - dose Zhidai Decoction), ( G5 ) vaginal group( vaginal filled with Zhidai Decoction), (G6)double azole oxazine group (vaginal filling with double azole oxazine). G2 to G6 were administered vagina with phenol glue for 3 d to build cervicitis model. And then, G1 to G6 were provided with the above- mentioned drugs. After 14 days,the body weight,pregnancy rate,the vaginal pathological score by naked eye, vagina and cervix HE staining, serum IL- 6/IL- 8 levels were detected. Results:After the modeling, compared with G1, the rats' vaginal pathological score, IL - 6 and IL - 8 were significantly increased (P 〈 0.05 ) in G2. Of course, the vagina and cervix' s pathological changes could be seen clearly, but the pregnancy rates were low. Compared with G2, the pregnancy rate increased in G3 to G6, while vaginal and cervical lesions relieved. In details, different doses and modes of administration had different efficacy. The pathological score, IL - 6 and IL - 8 were significantly lower ( P 〈 0.05 ) in G5. The IL - 6 and IL - 8 levels were significantly lower ( P 〈 0.05 ) in G3 and G6. Conclusion : Zhidai Decoction could reduce the pathological changes of cervicitis and increasing pregnancy rate. The underlying mechanism of improving the immune function may be related to reduced IL- 6 and IL- 8 levels.
