苍术中三种单体化合物对脾虚模型大鼠疗效研究

摘　　要：目的:通过研究苍术中三种单体化合物对痰湿困脾模型大鼠的疗效,来明确这3种化合物的药效。方法:以给大鼠灌服大剂量番泻叶为造模方法,再以脾虚模型大鼠为给药对象,分别给以不同剂量的苍术素、β-桉叶油醇以及5-羟甲基糠醛等,并测量不同组别大鼠的血清胃泌素(GAS)、血管活性肠肽(VIP)、一氧化氮(NO)、淀粉酶活性(AMS)、胃动素(MTL)等生理指标。结果:大鼠在灌服3种单体化合物后其胃泌素、血管活性肠肽、一氧化氮、淀粉酶活性、胃动素等生理指标均出现了不同程度的恢复,其中灌服高剂量5-羟甲基糠醛的大鼠血某些指标好于阳性药物组大鼠。结论:苍术素、β-桉叶油醇以及5-羟甲基糠醛这3种单体化合物均对脾虚模型大鼠有一定的治疗作用,提示这3种单体化合物可能是苍术健脾的物质基础。Objective ： To study the effects of three compounds separated from Atractylodes lancea （ Thunb. ） DC on spleen disorder rats to prove their curative effects. Methods ： The spleen disorder model was established by orally adminis- tered concentrated senna. And then the spleen disorder rats were orally administered Atractylodin, β - Eudesmol and 5 - hydroxymethylfurfural. The concentrations of GAS, VIP, NO, AMS and MTL in serum were measured by ELISA. The effects of AMS in serum were measured by Ultraviolet spectrophotometry. Results ： The concentration of GAS, VIP, NO and MTL in serum and the effect of AMS in serum of spleen disorder rats were quite different from the healthy one. Obviously after orally administered Atractylodin, β - Eudesmol and 5 - hydroxymethylfurfural, the detection indexes of spleen disorder rats recovered to be normal. The concentration of MTL in serum of the rats which orally administered 5 - hydroxymethylfurfural was higher than that of the positive drug group rats. Conclusions： Atractylodin, β- Eudesmol and 5 - hydroxymethylfurfural can treat spleen disorder rats and their curative effects are not as good as the water extract of Atractylodes lancea（ Thunb. ） DC but 5 - hydroxymethylfurfural in some detection indexes. The three compounds could be the reason for the Atractylodes lancea（Thunb. ）DC can treat spleen disorder.
