摘  要:目的:研究中药益糖康对糖尿病大鼠的疗效及其蛋白组学的变化,从蛋白质的分子水平上探索中药益糖康对糖尿病作用机制生物学基础。方法:高脂高糖饮食+用链脲佐菌素的方法建立糖尿病大鼠模型,给药10周后,检测血糖、血脂、胰岛素水平,并采用同位素标记技术,利用高效液相色谱仪及四级杆-静电场轨道阱质谱仪,对糖尿病大鼠进行串级质谱数据分析,对检测出的有统计学意义的表达差异白进行研究。结果:给药后,给药组与对照组比较,血糖从第6周开始有显著差异(P<0.01);治疗前后比较,给药组中的TC、TG、HDL-C有统计学意义(P<0.01),LDL-C无显著差异(P>0.05);对照组中所有指标均无统计学差异(P>0.05);组间比较,给药组的TC、TG、HDL-C、胰岛素(INS)、胰岛素敏感指数(IAI)、胰岛素抵抗指数(IR)改善明显优于对照组(P<0.01),LDL-C无显著差异(P>0.05);蛋白组学变化,给药组与对照组比较,共得到40个差异蛋白,其中上调大于1.5倍的蛋白有11个,下调小于0.75倍的有29个。结论:中药益糖康是通过改善和修复蛋白的合成而降低血糖,并改善糖脂代谢的,从蛋白质分子水平上发现了中药益糖康治疗糖尿病作用机制的生物学基础。Objective: This research was based on the effect of Chinese medicine Yitangkang on diabetic rats and the change of proteomics to explore the treatment mechanism of biological basis of Yitangkang from protein molecules. Methods: The diabetic rat model was established by using the method of the high fat and high glucose diet + streptozotocin (STZ). For ten weeks of drug treatment, the experiment monitored blood glucose, blood lipid,insulin levels. And the serial spectrometry data analysis of diabetic rats was obtained with the isotope labeling technique and high performance liquid chromatography and four bar - orbitrap mass spectrometer techniques and then the statistically significant difference in protein expression was measured. Results: After treatment with drugs, the blood glucose started to appear significant differences when comparing drug group with control group at the sixth month. Before and after treatment ,TC ,TG and HDL -C were statistically significant in the drug group( P 〈 0. 01 ) and LDL- C (P 〉 0. 05 )was not. All indicators were not statistically significant in the control group. TC,TG,HDL - C,INS,IAI and IR of the drug group were much better than those of the control group ( P 〈 0. 01 ) and LDL - C ( P 〉 0. 05 ) was not. From the proteomic changing, proteomics detection obtained 40 differential proteins,in which, 11 proteins were up - regulated more than 1.5 times, and 29 proteins down - regulated less than 0.75 times. Conclusion: This research shows that Chinese medicine Yitangkang was able to lower glucose and improve the metabolism glucose and lipid through the improvement and repair protein synthesis. This research find out the mechanism of biological basis that Yitangkang cures the diabetes from protein molecules.
