摘  要:目的:研究紫檀芪(pterostilbene,PTE)体外抑制C6胶质瘤细胞增殖并诱导其凋亡的作用,探讨其可能的作用机制。方法:运用MTT法测定细胞存活率,吖啶橙/溴化乙锭(AO/EB)双荧光染色观察C6细胞凋亡形态学变化,酶联免疫吸附法(ELISA)测定Caspase-3活性变化,并用Western blot法检测Fas、FasL蛋白的表达水平。结果:由MTT结果知紫檀芪可以显著抑制C6细胞的增殖(P<0.01),呈现药物浓度与时间依赖性;AO/EB荧光染色(24 h)可见典型的凋亡形态学特征;酶联免疫吸附(ELISA)检测表现为随PTE浓度升高,Caspase-3活性逐步升高(P<0.01);Western blot检测显示PTE可以使Fas、FasL蛋白表达显著增加(P<0.01)。结论:PTE可诱导胶质瘤细胞系C6细胞凋亡,其作用机制可能与激活Fas/FasL通路、上调Caspase-3活性有关。Objective:To investigate the effect of pterostilbene(PTE) on the proliferation and apoptosis of glioma cell C6 in vitro,to probe into its mechanism.Method:MTT method was used to detect the survival rate of cells,using AO/EB fluorescent staining methods to observe apoptotic morphological changes of C6 cells.The enzymatic acitivity of Csapase-3 was measured by ELISA.The protein levels of Fas and FasL were detected by Western blot.Result:MTT result showed that PTE can effectively inhibit the proliferation of C6(P〈0.01),these effects were showed to be time-and dose-dependent.The result of AO/EB fluorescent staining(24 h) showed that C6 treated by PTE was seen typical apoptotic morphological features.Besides,the enzymatic activity of Csapase-3 was promoted obviously after the effect of PTE on C6 cells by ELISA(P〈0.01).The result of Western blot showed that the protein levels of Fas and FasL in the C6 were upgraded significantly(P〈0.01). Conclusion:PTE can induce apoptosis of glioma cell C6,which is concerned with upgrading Fas and FasL,thereby affecting activation of apoptotic factor Caspase-3 protein.
