摘　　要：目的研究Necrostatin-1(Nec-1)对大鼠心肌梗死后心梗面积的影响。方法成年雄性SD大鼠共25只,随机分为Nec-1处理组(实验组,10例)、安慰剂对照组(对照组,10例)和假手术组(5例);通过结扎冠状动脉左前降支建立心肌梗死模型,并于结扎的同时,给予大鼠尾静脉Nec-1(0.6 mg/kg)或相应溶剂对照;在缺血2周后,处死大鼠,取出心脏,用Masson氏染色法观察并测定相应的心肌梗死面积。结果实验组大鼠心梗面积比对照组大鼠显著减少,差异有统计学意义(P<0.01)。结论 Nec-1可明显地抑制心肌缺血大鼠的心肌坏死,有确切的心肌保护作用。Objective To study the effect of Necrostatin-1( Nec-1) on myocardial infarct size in rats with myocardial infarction. Methods Twenty-five adult male Sprague-Dawley rats were randomly divided into Nec-1 treated group( experimental group,n = 10),placebo control group( control group,n = 10) and sham operation group( n = 5). The myocardial infarction model was established by ligating the left anterior descending coronary artery( LAD). At the same time,the rat tail vein Nec-1( 0. 6 mg/kg) or the corresponding solvent control was given. After two weeks of ischemia,the rats were sacrificed and the hearts were removed. Masson stain was used to measure the infarct size. Results Compared with the control group,the myocardial infarct size of experimental group,was significantly decreased( P < 0. 01). Conclusion Nec-1 can significantly inhibit the myocardial necrosis in rats with myocardial ischemia,and has a definite myocardial protective effect.
