摘 要:目的探讨益母草碱对慢性心力衰竭心肌重构大鼠血清心房利钠肽(atrial natruretic peptide,ANP)、血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)的影响。方法 10只正常SD大鼠作对照组,腹腔注射生理盐水;另80只SD大鼠作为实验鼠,连续2周腹腔注射异丙肾上腺素诱导慢性心力衰竭动物模型。造模成功后实验鼠随机分为5组:模型组(生理盐水)、益母草碱低剂量组(给益母草碱量为7.5mg/kg/d)、中剂量组(给益母草碱量为15mg/kg/d)、高剂量组(给益母草碱量为30mg/kg/d)、p38丝裂原活化蛋白激酶(mitogen-activated protein kinases,P38MAPK)抑制剂组(给P38MAPK量为0.3mg/kg/d),连续予相应药物腹腔注射2周后行心脏超声检测心脏结构及功能,ELIAS法检测血清ANP、AngⅡ含量。结果(1)与正常组比较,模型大鼠左室后壁厚度(LVPW)、室间隔厚度(IVS)、左室舒张末期内经(LVDd)、左室收缩末期內径(LVDs)以及血清ANP、AngⅡ含量明显增加,而左室射血分数(EF%)、左室短轴缩短率(FS%)则明显降低,差异均有统计学意义(P<0.05)。(2)与模型组比较,益母草碱高剂量组和P38MAPK抑制剂组LVPW、IVS、LVDd、LVDs及血清ANP、AngⅡ含量明显降低,EF%、FS%则明显升高,差异均有统计学意义(P<0.05)。结论高剂量益母草碱(30mg/kg/d)通过减少慢性心力衰竭大鼠血清ANP、AngⅡ含量对抑制心肌重构有一定作用。Objective To investigate the effects of Leonurine on serum BNP and Ang II in the myocardial remodeling of rat with chronic heart failure.Methods 10 normal SD rats as control group,meanwhile 80 experimental rats were given isoproterenol of intraperitoneal injection daily for 2w,then the rats with chronic heart failure were divided randomly into five groups,which were the model group,low-dose Leonurine group(7.5mg/kg/d),middle-dose Leonurine group(15mg/kg/d),high-dose Leonurine group(30mg/kg/d),and P38 MAPK inhibitor group.During the experiment,different drugs and dose were intraperitoneal injections to different groups and the normal saline was used for normal group and model group.After 2w,cardiac structure and function was tested by echocardiography,content of serum ANP and Ang II were detected by ELISA respectively.Results Compared with those in normal group,LVPW,IVS,LVDd,LVDs,the levels of serum ANP and Ang II in model group were progressively increased,but EF% and FS% were significantly decreased(all P〈0.05).Compared with those in model group,LVPW,IVS,LVDd,LVDs,the levels of serum ANP and Ang II in high-dose Leonurine group and P38 MAPK inhibitor group were progressively decreased,but EF% and FS% were significantly increased(all P〈0.05).Conclusion High-dose Leonurine group(30mg/kg/d)can inhibit myocardial remodeling in the rat with chronic heart failure maybe through attenuating serum BNP and Ang II.
