龙胆泻肝丸中3种活性成分的HPLC-DAD法测定

摘　　要：目的:分析HPLC-DAD法测定对龙胆泻肝丸中3种活性成分(黄芩苷、栀子苷、龙胆苦苷)的测定情况。方法:选用龙胆泻肝丸、及3种活性成分对照品进行HPLC-DAD法检测。色谱柱:PLATSILTMODS,250 mm×4.6 mm,5μm,色谱柱的柱温为30℃,流速:1 m L/min。分别对检测药物进行梯度洗脱,内标液为甲醇、0.1%磷酸水溶液,黄芩苷和龙胆苦苷的检验波长为280 nm,栀子苷的检验波长为240 nm。结果:经过HPLC-DAD法测定后,黄芩苷的线性范围为0.292-5.290μg(r=0.9998),平均回收率为100.2%;栀子苷的线性范围为0.0077-0.1366μg(r=0.9999),平均回收率为99.6%;龙胆苦苷的线性范围为0.111-2.018μg(r=0.9999),平均回收率为102.2%。3种活性成分的平均回收率均超过99%。结论:HPLC-DAD法是一种新型检测方法,操作简单、准确性和稳定性高,重复性强,适用于药动学研究及监控龙胆泻肝丸的生产质量,保证用药安全。Objective ： To analyzethe determination of three kinds of active ingredients （ baicalin, geniposide, gentian bitter gly- coside） in Longdan Xiegan Pill by HPLC -DAD method. Methods：Use Longdan Xiegan Pilland three kinds of active ingredients reference substance to do the HPLC - DADtest. Chromatographic column ： PLATSILTM ODS ,250 mm × 4.6 mm,5 microns, chromatographic column column temperature 30 ℃, velocity ： 1 mL/min. Respectively to gradient elution of testing drugs, internal standard solution for methanol ,0.1% phosphoric acid solution, baicalin and gentian bitter glycosides inspection wavelength of 280 nm, gardenoside test wavelength of 240 nm. Results ： After HPLC - DAD method,linear range was 0. 292 to 0. 292 μg of baicalin （r = 0. 9998 ） and the average recovery was 100.2%. Geniposide linear was in the range of 0.0077 to 0. 1366 μg （r = 0. 9999） and the average recovery was 99.6%. Gentian bitter glycosides linear was in the range of 0.111 to 2. 018 μg（ r = 0. 9999 ） and the average recovery was 102.2%. The average recovery of three kinds of active ingredients was more than 99%. Conclusion： HPLC -DAD method is a new kind of detection method, simply operated, with high accuracy and stability. The repeatability is strong, suitable for pharmaeokinetic studies and monitoring the production quality of Longdan Xiegan Pill, ensuring the safety of medication.
