摘 要:目的探索铁皮石斛对二甲基肼(1,2-dimethylhydrazine,DMH)诱发小鼠结直肠癌(colorectal cancer,CRC)的预防作用。方法于2014年5月,将90只ICR小鼠随机分为正常对照组、单纯铁皮石斛组、模型组,低、中、高剂量铁皮石斛预防组。模型组和预防组以DMH 20mg/kg腹腔注射,每周1次,各预防组分别喂饲含5%、10%和20%铁皮石斛的饲料。观察动物的一般表现,检测血清超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA),检查肿瘤发生率,进行组织病理学检查。结果中、高剂量预防组的体重[(47.7±1.5)、(46.8±2.8)g]和血清SOD水平[(179.14±14.09)、(179.22±8.05)U/L]均明显高于模型组(P<0.05),结直肠癌发生率(40.00%、26.67%)和结直肠黏膜腺体增生程度(中、轻度增生)均明显低于模型组(P<0.05)。结论在本实验条件下,铁皮石斛对于DMH所诱发的CRC具有一定的预防作用,铁皮石斛应用于预防肿瘤具有较广阔的前景。Objective To explore the preventive effect of Dendrobium officinale on colorectal cancer(CRC)induced by1,2-dimethylhydrazine(DMH).Methods A total of 90 ICR mice were divided randomly into the normal control group,the simple Dendrobium officinale group,the model group,prevention groups with low,medium and high dose of Dendrobium officinale in May 2014.The mice were injected intraperitoneally with DMH at the dose of 20mg/kg body weight once a week in the model group and the prevention groups.The mice were fed with 5%,10% and 20% Dendrobium officinale in the prevention groups,respectively.The general information of mice was observed.The serum SOD and MDA were detected.The CRC incidence were investigated.The histopathological examinations were carried out.Results The body weight was(47.7±1.5)and(46.8±2.8)g,the serum SOD level was(179.14±14.09)and(179.22±8.05)U/L in the prevention group with medium,high dose of Dendrobium officinale respectively.They were significantly higher than those in the model group(P 0.05).The CRC incidence was 40.00% and 26.67%,the degree of glandular hyperplasia in colorectal mucosa was moderate and mild in the prevention group with medium,high dose of Dendrobium officinale respectively.They were significantly lower than those in the model group(P 0.05).Conclusion Under the experimental conditions,Dendrobium officinale has a good preventive effect on CRC induced by DMH.There is a broad prospect that Dendrobium officinale be used to prevent cancer.
