摘 要:目的:观察黄连素对高脂饲料诱导高脂血症小鼠脂质代谢与抗氧化水平的影响。方法:50只雄性昆明小鼠,随机分为对照组、模型组和黄连素低、中、高剂量组,每组10只。除了对照组饲喂基础饲料,其余四组均采用高脂日粮构建小鼠高血脂模型,建模同时,黄连素低、中、高剂量组分别灌胃50、100、200mg/(kg·d)进行干预,7周后测定小鼠血浆甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDLch)、高密度脂蛋白胆固醇(HDL-ch)、总蛋白(TP)、白蛋白(ALB)、丙二醛(MDA)含量以及丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)活力,并计算动脉硬化指数、肝脏与腹内脂肪相对重。结果:与模型组相比,黄连素可降低高脂日粮诱导的高脂血症小鼠的血脂水平、动脉硬化指数、MDA含量、血浆ALT和AST活性,提高血浆SOD活性、血浆TP、ALB浓度,同时降低肝脏和腹内脂肪相对重。结论:结果显示黄连素可以调节高脂血症模型小鼠的脂质代谢及其导致的动脉粥样硬化与肝损伤,同时具有一定的抵抗脂质过氧化作用。Objective:To explore the effect of berberine on lipid metabolism and antioxidant ability in hyperlipidemic mice induced by high fat feed. Methods:Fifty Kunming male mice were randomly divided into 5 groups,the normal control group,the model group,and the berberine low,middle,high dose group,10 in each group.Except those in the normal control group,all mice were fed with high fat feed to prepare hyperlipidemic mice model,meanwhile,these mice were given intragastrically berberine of 0,50,100,200 mg/kg for 7 weeks. The plasma levels of total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-ch),high-density lipoprotein cholesterol(HDL-ch),total protein(TP),albumin(ALB),alanine aminotransferase(ALT),aspartate aminotransferase(AST),superoxide dismutase(SOD) and malondialdehyde(MDA)were measured,and atherogenic index(AI),the relative weight of liver and abdominal fat were evaluated. Results:Compared with the model group,the three groups treated with berberine exhibited significantly lowered the plasma levels of TC,TG,LDL-ch,ALT,AST and MDA,elevated plasma SOD activity and HDL-ch,TP and ALB content,and reduced significantly AI,the relative weight of liver and abdominal fat. Conclusion:It was indicated that berberine could regulate lipid metabolism,atherosderosis and liver damage,and had a certain resisting lipid peroxidation in hyperlipidemic mice.
