摘 要:应用通用引物测定了观赏鱼市场上具有较高价值的7个品系七彩神仙鱼和3个品系神仙鱼的16S r RNA基因和CO I基因部分序列,并应用DNA序列分析软件,对这些基因进行了同源性比较和系统发育分析。通过Gen Bank中的核酸BLAST和BOLD SYSTEMS数据库对这些基因序列进行比对,结果表明16S r RNA基因和CO I基因可以在物种水平对神仙鱼进行准确鉴定,但对七彩神仙鱼只能进行有限度的鉴别。由于不同品系的七彩神仙鱼或神仙鱼,尽管其体色和斑纹具有明显差异,但16S r RNA基因和CO I基因序列高度相似。因此,对这些观赏鱼依靠16S r RNA基因或CO I基因都无法准确鉴定,需要研究进化速度更快的DNA基因条码。The discuses(Symphysodon sp) and angelfish(Pterophyllum sp) were ornamental fishes which had high economic value. Partial sequence of 16 S r RNA gene and mitochondrial cytochrome oxidase subunit I(CO I) gene from 7 lines of discuses and 3 of angelfish which had higher value in the market had been amplified using universal primers. The homology and phylogenetic analysis of these genes were carried out by using DNA sequence analysis software. The gene sequence had been compared with the database of Gen Bank and BOLD SYSTEMS. The phylogenetic tree had been established based on the16 S r RNA gene and the CO I gene. Sequence analysis showed that the 16 S r RNA and CO I genes can be used to identify the angelfish accurately in specie level,but it had only limited function in discrimination for discuses. Because of the high similarity of sequences between the different lines of discuses or angelfish,although it had obvious difference in body color and stripes,it is difficult to identify these fishes based on the 16 S r RNA or CO I gene. The DNA barcode which had higher rate of evolution should be studied.
