摘 要:目的:探讨补肾中药复方首参颗粒对ApoE^(-/-)小鼠动脉粥样硬化斑块的影响及可能机制。方法:将72只Apo E-/-小鼠随机分为空白对照组(空白组)、模型组、首参颗粒组、普伐他汀组,每组18只。空白组喂饲普通饲料,其他各组喂饲高脂饮食。空白组和模型组小鼠予双蒸水灌胃,首参颗粒组给予首参颗粒水溶剂灌胃(每日生药量8 g/kg),普伐他汀组予普伐他汀水溶剂灌胃(每日1.8 mg/kg),持续12周。12周后,HE染色观察主动脉根部病理变化;Real-time PCR法检测主动脉p66^(Shc)mRNA水平;免疫组化和Western blot法检测小鼠主动脉p66^(Shc)蛋白表达。结果:模型组小鼠动脉内有明显AS斑块形成,斑块面积明显大于空白组,差异有统计学意义(P<0.05);与模型组比较,首参颗粒组、普伐他汀组斑块面积明显减少(P<0.05)。模型组小鼠主动脉p66^(Shc)的mRNA表达及蛋白表达水平明显高于空白组;与模型组比较,首参颗粒组、普伐他汀组主动脉p66^(Shc)的mRNA表达和蛋白表达水平明显降低(P<0.01)。结论:补肾中药复方首参颗粒可改善动脉粥样硬化动物的斑块形成,其机制可能与降低主动脉p66^(Shc)mRNA表达和蛋白表达有关。Objective: To discuss the effects and possible mechanism of Shoushen Granule on atherosclerotic plaque in ApoE -/- mice. Methods: 72 ApoE-/- mice were randomly divided into the blank control group, model group, Shoushen Granule group and Pravastatin group, 18 in each group. The blank group was fed with normal diet, and the other groups were fed with high fat diet. The blank control group and model group were treated with double distilled water for intragastric administration. The Shoushen Granule group was treated with water solvent of Shoushen Granule at dose of 8 g crude drug/kg daily. The Pravastatin group was treated with water solvent of Pravastatin at dose of 1.8 mg/kg daily. All the treatment course were 12 weeks. The pathological changes of aortic root were observed by HE stain. The p66Shc mRNA level in aorta was detected by Real-time PCR. The protein expression of p66shc in aorta was detected by immunohistochemistry and Western blot. Results: Significant atherosclerotic plaque was found in the artery of mice in the model group, the plaque area was significantly larger than that in the blank group, with statistically significant difference (P 〈 0.05 ). The plaque areas in the Shoushen Granule group and Pravastatin group were significantly decreased compared with the model group ( P 〈 0.05 ). The mRNA and protein expression levels of p66shc in aorta of mice in the model group were obviously higher than those in the blank control group. The mRNA and protein expression levels of p66shc in the Shoushen Granule group and Pravastatin group were significantly decreased compared with the model group ( P 〈 0.01 ). Conclusion : Shoushen Granule can improve the plaque formation in atherosclerotic animals, and its mechanism may be related to decreasing the mRNA and protein expression of p66shc in aorta.
