摘 要:目的:探讨柴郁地仙方抗围绝经期抑郁症的作用途径。方法:40只SD大鼠分为假手术组、模型组、盐酸氟西汀组、柴郁地仙方组,每组10只。假手术组仅切除部分脂肪组织,余行双侧卵巢去势后予以21 d慢性轻度不可预见性应激(chronic unpredictable mild stress,CUMS)建立抑郁症动物模型,各组在CUMS第1天起每日分别以生理盐水、盐酸氟西汀及柴郁地仙方灌胃,持续21 d。观察刺激过程大鼠行为学变化,实验结束后断头取脑,采用酶联免疫吸附测定(enzyme linked immunosorbent assay,Elisa)法测定下丘脑5-羟色胺(5-hydroxytryptamine,5-HT)、多巴胺(dopamine,DA)、去甲肾上腺素(norepinephrine,NE)水平。结果:柴郁地仙方具有维持大鼠Open-Field得分、24 h糖水消耗量稳定、保证其体质量正常增长、上调下丘脑5-HT、NE、DA水平的作用,与模型组比较差异有统计学意义(P<0.01)。结论:柴郁地仙方抗围绝经期抑郁症作用途径与改善大鼠行为学、影响下丘脑神经递质分泌相关。Objective: To study the mechanism of Chaiyu Dixian Prescription (CYDXP) on perimenopausal depressive disorder (PDD). Methods: 40 SD rats were divided into the sham-operation group, model group, fluoxctine hydroehloride group and CYDXP group, 10 in each group. The sham-operation group was received sham operation. After ovariectomized surgery, the PDD model was established by chronic unpredictable mild stress (CUMS) for 21 days. From the first day of CUMS, each group was intragastrically treated with normal saline, fluoxetine hydrochloride and Chaiyu Dixian Prescription for 21 days, respectively. The behavioral changes of rats in the stimulation process were observed. After experiment, the brain tissue was collected. The levels of 5-hydroxytryptamine ( 5-HT ) , dopamine ( DA ) and norepinephrine ( NE ) in hypothalamus were determined by enzyme linked immunosorbent assay(ELISA). Results: Chaiyu Dixian Prescription could maintain the Open- Field score, stabilize the 24 h sucrose consumption, ensure the normal growth of body weight, up-regulate the levels of 5-HT, NE and DA. The differences were statistically significant compared with the model group( P 〈 0.01 ). Conclusion: The inhibition effects of Chaiyu Dixian Prescription on perimenopausal depressive disorder may be related to the improvement of rats ' behaviour and the regulation of neurotransmitter secretion in hypothalamus.
