摘  要:目的:制备不同组方的中药药物性口罩,并评价其对流感病毒H1N1的抑制作用。方法:组方1(射干、甘草)与组方2(金银花、甘草)分别乙醇提取,以提取液浸泡纯棉脱脂纱布口罩2 h,分别制备1号、2号口罩,烘干备用。用制备药物性口罩的原液作用于流感病毒感染的犬肾细胞(MDCK),取细胞上清液进行血凝试验,比较各组细胞上清液的血凝效价,Real-time PCR法测定细胞内流感病毒血凝素(HA)基因的mRNA表达;将流感病毒H1N1液分别过滤药物性口罩,收集口罩滤液,检测滤液中流感病毒HA的效价。结果:组方1作用于流感病毒感染的MDCK细胞后,上清液的血凝效价由210降为26,组方2的血凝效价由210下降为25;与模型组比较,药物组细胞内流感病毒HA基因的mRNA表达量均明显减少(P<0.05)。1号口罩过滤流感病毒H1N1液后,HA的效价由2-12下降为2-5,2号口罩过滤后的效价由2-12下降为2-6。结论:中药组方醇提液可有效抑制流感病毒在MDCK内RNA的复制;中药药物性口罩能够有效破坏流感病毒包膜上的血凝素,可有效阻断流感病毒的传染。Objective: To prepare the breathing mask of traditional Chinese medicine with different prescriptions and evaluate the inhibitory effects on influenza virus H1N1. Methods: Prescription No. 1 (Belameanda ehinensis and Glycyrrhiza uralensis) and prescription No. 2 (Lonicerae japonicae flos and Glyeyrrhiza uralensis )were extracted by ethanol respectively. The pure cotton absorbent gauze mask was soaked in the extract liquid for 2 hours, and the breathing mask No. 1 and No. 2 were prepared after drying. Madin-Darby canine kidney (MDCK) cells infected by influenza virus were intervened by the original solution which used for the mask preparation. The cell supernatant was collected and the hcmagglutination assay was carried out,the potency of hemagglutination of cell supernatant in each group was compared. The intracellular mRNA expression of influenza virus hemagglutinin (HA) gene was detected by RT-PCR. Different masks were used to filter the influenza virus H1 N1 solution and the filtrate was collected and the potency of influenza virus HA in the filtrate was determined. Results: After the intervention of prescription No. 1 on influenza virus-infected MDCK cells, the potency of hemagglutination of cell supematant was decreased from 210 to 26 , after the intervention of prescription No. 2, the potency of hemagglutination of cell supernatant was decreased from 210 to 25. Compared with the model group, the intraceflular mRNA expression of influenza virus HA gene was obviously decreased in the drug treatment groups (P 〈 0.05 ). After the filtration of influenza virus H1 N1 solution by mask No. 1, the potency of HA was decreased from 2- 12 to 2 -5 ;after the filtration by mask No. 2, the potency of HA was decreased from 2 -12 to 2 -6. Conclusion: The alcohol extract of traditional Chinese medicine prescription can effectively inhibit the RNA replication of influenza virus in MDCK.
