摘 要:对6省9个不同地区川芎种质、藁本及辽藁本样品的ITS2序列进行分析,并采用NJ法构建分子系统树。结果显示:9个川芎种质、藁本与辽藁本的ITS2序列共有220个保守位点和8个变异位点,其中信息位点4个,单一变异位点4个。藁本、辽藁本与川芎间GC含量相近,变异位点相似,平均K2P遗传距离分别为0.008,0.011,小于川芎种内遗传距离0.013。NJ进化树分析显示,各地川芎种质、藁本、辽藁本间有着极为亲密的亲缘关系。结合前人本草考证的结果,对川芎的起源、川芎种质间的亲缘关系进行了讨论,首次提出辽藁本也可能是非主栽川芎种质野生种源的观点。ITS2sequences of L.sinensecv.‘Chuanxiong',L.sinense Oliv.and L.jeholensefrom6 provinces and 9regions were amplified and sequenced,and the phylogenetic tree was constructed by neighbor-joining(NJ).The results showed that there were 220 conserved sites and8 variable sites in the ITS2 region.The GC frequency and the mutation sites of ITS2 sequence of L.sinense cv.‘Chuanxiong'germplasms L.sinense Oliv.and L.jeholense from different regions,were similar.The K2 Pdistances between L.sinense cv.‘Chuanxiong'and L.sinense Oliv.as well as that between L.sinense cv.‘Chuanxiong'and L.jeholense were 0.008and0.011,respectively,which were less than the intraspecies genetic distance of L.sinense cv.‘Chuanxiong',0.013.The NJ phylogenetic trees suggested that dose relationship was found among L.sinense cv.‘Chuanxiong'germplasms,L.sinense Oliv.,and L.jeholense.Based on researches on Materia Medica,the origin of L.sinense cv.‘Chuanxiong',and the relationship among L.sinense cv.‘Chuanxiong'germplasms were discussed.The viewpoint that L.jeholense may be the origin of non-mainstream cultivated varieties of L.sinensecv.‘Chuanxiong'was put forward for the first time.
