摘 要:目的:探讨母源性丙烯酰胺(acrylamide,ACR)暴露对断乳期大鼠额叶皮质生长相关蛋白(GAP-43)和脑源性神经营养因子(BDNF)表达的影响。方法:SD孕鼠随机分为4组,怀孕第6 d起对照组和实验组分别给予双蒸水和50,100,200μg/ml ACR饮水,直至仔鼠出生21 d,染毒结束时对断乳期大鼠称重。免疫组织化学检测额叶皮质GAP-43和BDNF的表达。结果:与对照组相比,实验各组断乳期大鼠体重呈浓度依赖性下降,且组间差异明显(P<0.05)。与对照组相比,ACR实验各组GAP-43表达显著降低(P<0.05),而低、中剂量组之间差异不显著(P>0.05),高剂量组与低、中剂量组相比,表达显著减少(P<0.05);与对照组相比,ACR实验组各组BDNF表达显著减少(P<0.05),中、高剂量组与低剂量组相比,BDNF表达减少(P<0.05),而中、高剂量组之间差异不显著(P>0.05)。结论:母源性ACR染毒可能通过下调额叶皮质神经元GAP-43和BD)NF表达,抑制轴突延伸或扰乱神经元发育的内环境稳态,进而影响额叶皮质的发育。Objective;To explore the influence of maternal acrylamide exposure on the expression of growth associated protein(GAP-43) and brain derived neurotrophic factors(BDNF) in the cerebral frontal cortex of weaning rats.Methods:The SD pregnant rats were randomly divided into 4 groups and provided with doubly evaporated water in control groups and50,100,200 μg/ml ACR solution via drinking water in experimental groups respectively from gestational day 6 to postnatal day 21 and weighed the weaning rats at the end of the experiment.The expression of GAP-43 and BDNF was observed in the weaning rats frontal cortex neurons by immunohistochemistry.Results:Compared with the control group,the body weight of weaning rats in the ACR experimental groups decreasedsignificantly in a dose dependent manner(P〈0.05),and the difference between ACR experimental groups was also significant P〈0.05).Compared with the control group,the expression of GAP-43 in each ACR experimental group decreased significantly(P〈0.05),while the expression of GAP-43 between the low-dose group and mid-dose group had no statistical significance(P〉0.05),the expression of CAP-43 in high-dose group decreased significantly than that of the low-dose group and mid-dose group(P〈0.05);Compared with the control group,the expression of BDNF in each ACR experimental group were lower than that of the control group,this index of the mid-dose group and high-dose group is much more lower than that of the low-dose group(P〈0.05),while there were no significant difference between the mid-dose group and high-dose group(P〈0.05).Conclusion:Maternal acrylamide exposure might inhibit axonal plasticity and interfere with homeostasis by decreasing the expression of CAP-43 and BDNF,which then may affect the development of frontal cortex.
