糖尿病大鼠肾脏及血清炎症因子的表达

摘　　要：目的探讨血管内皮生长因子(VEGF)、细胞间黏附分子1(ICAM-1)、TNF-α、IL-6和IL-1β在糖尿病大鼠肾脏及血清中的表达差异。方法采用链脲佐菌素诱导27只SD大鼠成功建立糖尿病模型(模型组),分别于成模后2周、2个月采用ELISA法检测肾脏组织及血清中VEGF、TNF-α、ICAM-1、IL-1β和IL-6的含量。另取10只SD大鼠作为对照组,常规饲养2个月取血清及肾脏组织行上述炎症因子的检测。结果模型组成模后2个月,大鼠血清VEGF、TNF-α、ICAM-1、IL-1β和IL-6表达均高于对照组(P<0.05或P<0.01)。模型组成模后2周和2个月,大鼠肾脏中VEGF、TNF-α、ICAM-1、IL-1β和IL-6表达均高于对照组(P<0.05)。模型组成模后2个月,大鼠肾脏中VEGF、IL-1β和IL-6表达高于成模后2周(P<0.05),而TNF-α和ICAM-1表达差异无统计学意义(P>0.05)。结论糖尿病大鼠肾脏及血清中炎症因子VEGF、TNF-α、ICAM-1、IL-1β和IL-6表达均明显增加,且肾脏中VEGF、IL-1β和IL-6表达随病程延长而增加。Objective To explore the expressions of vascular endothelial growth factor（VEGF）,intercellular adhesive molecular-1（ICAM-1）,tumor necrosis factor-α（TNF-α）,IL-6and IL-1βin the kidney and serum of diabetic rats.Methods The diabetic rat models were induced by streptozotocin successfully in 27 male SD rats（group A）.The expressions of VEGF,ICAM-1,TNF-α,IL-6and IL-1βin the kidney and serum were detected by ELISA in 2 weeks and 2 months after establishment of diabetic model.Another 10 SD rats（group B）were taken as the controls.Results Serum levels of VEGF,TNF-β,ICAM-1,IL-1βand IL-6were higher in group A than those in group B in 2months after model establishment（P〈0.05 or P〈0.01）.The expressions of VEGF,TNF-α,ICAM-1,IL-1βand IL-6were higher in the kidney tissues of group A were higher than those in group B in 2months and 2weaks after model establishment.（P〈0.05）.The expressions of VEGF,IL-1β,and IL-6,but not TNF-αand ICAM-1,in the kidney tissues of group A were higher in 2months than those in 2 weeks after models were established（P〈0.05）.Conclusion The expressions of VEGF,TNF-α,ICAM-1,IL-1βand IL-6in the kidney and serum of diabetic rats are increased.The expressions of VEGF,IL-1βand IL-6in the kidney increase as diabetic course prolongs.
